摘要
目的观察红豆杉多糖对D-半乳糖衰老小鼠模型肝功能的影响,并对其作用机制进行初步探讨。方法90只雄性ICR小鼠,随机挑选15只作为空白组,其余75只建立衰老模型,随机分为模型组(D-半乳糖)、阳性药物组(易善复)以及红豆杉多糖低剂量组、中剂量组、高剂量组,每组各15只。造模第4周起,阳性药物组给予多烯磷脂酰胆碱(易善复)干预,红豆杉多糖低、中、高剂量组分别使用不同浓度红豆杉多糖溶液灌胃干预;空白组和模型组使用等剂量生理盐水灌胃苏木精-伊红染色观察肝脏组织病理学改变,比色法比较肝组织匀浆中总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)活性以及丙二醛(MDA)含量,全自动生化仪检测血清谷草转氨酶(AST)、谷丙转氨酶(ALT)含量。结果模型组的SOD活性、T-AOC较空白组明显下降,MDA含量、ALT及AST含量较空白组明显上升,差异有统计学意义(P<0.01)红豆杉多糖低/中/高剂量组和阳性药物组的SOD活性、T-AOC较模型组明显升高,MDA含量、ALT及AST含量较模型组明显降低,差异有统计学意义(P<0.05)结论红豆杉多糖可能通过提高抗氧化应激能力而改善D-半乳糖所致的衰老小鼠模型肝功能损害。
Objective To explore the effect and mechanism of Taxus polysaccharide on the liver function of aging mice induced by D-galactose.Methods Fifteen of the 90 male ICR mice were randomly selected as blank group,and the remaining 75 mice were randomly divided into model group(D-galactose),positive drug group(Essentiale N),taxiis polysaccharide low-dose group,medium-dose group and high-dose group,with 15 mice in each group.From the 4th week of modeling,the positive drug group was given polyene phosphatidylcholine(Essentiale N),and the low,medium and high dose taxus polysaccharide groups were given different concentrations of taxus polysaccharide solution by intragastric intervention.The blank group and model group were given equal dose of nomial saline intragastric administration.Observe the histopathological changes of liver by HE staining.The total antioxidant capacity(T-AOC),the activity of superoxide dismutase(SOD)and the level of malondialdehyde(MDA)of liver homogenate were detected by colorimetry.The level of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum of mice were detected by automatic biochemical analyzer.Results The activity of SOD and T-AOC in the model group were significantly decreased compared with those in the blank group,while the content of MDA,ALT and AST in the model group were significantly increased compared with those in the blank group,the difference was statistically significant(P<0.01).The activity of SOD and T-AOC in taxus polysaccharide dosage groups and positive drug group were increased compared with those in the model group,while the levels of MDA,ALT and AST in different doses of taxus polysaccharide groups were significantly decreased,the difference was statistically significant(P<0.05).Conclusion Taxus polysaccharide may improve the liver function damage induced by D-galactose in aging mice models through antioxidant stress damage.
