清溪花鳖肽对尿酸钠所致大鼠痛风性关节炎的作用

Effect of Qingxi Softshell Turtle Peptides on Gout Arthritis in Rats Caused by Sodium Urate

清溪花鳖肽(QXSTP)是浙江清溪花鳖中蛋白质的酶法水解产物,本研究将探讨QXSTP治疗尿酸钠所致大鼠痛风性关节炎的作用,为进一步开发该产品提供试验依据。将雄性SD大鼠随机分成空白对照组、模型对照组、秋水仙碱对照组、痛风舒片对照组、QXSTP高、中、低剂量组,每天给药1次,连续给药28 d。d 26给药后30 min,除空白对照组外,其余各组尿酸钠溶液注入大鼠踝关节腔,造大鼠痛风性关节炎模型,分别进行踝关节肿胀度检测、步态评分、血清尿素(UA)、TNF-α和IL-6含量测定及组织病理学检测。与模型对照组相比,造模后12和24 h,QXSTP低、中、高剂量组大鼠踝关节肿胀度均有不同程度的降低;造模后24 h,QXSTP中、高剂量组大鼠步态评分显著降低;QXSTP中、高剂量组大鼠血清中UA和TNF-α含量有不同程度的降低,高剂量组大鼠血清中IL-6的含量极显著降低;与模型对照组相比,QXSTP低、中、高剂量预防给药均能显著降低HE染色病理评分。在本实验条件下,QXSTP对尿酸钠所致大鼠痛风性关节炎有一定的治疗作用。

Qingxi Softshell Turtle Peptides(QXSTP)are enzymatic hydrolysate of proteins in Zhejiang Qingxi Softshell Turtle.The authors'study is to investigate the effect of QXSTP on gouty arthritis induced by sodium urate in rats,as well as providing a test basis for the further development of QXSTP.Healthy male SD rats were randomly divided into blank control group,model group,colchicine group,Tongfengshu group,high/medium and low dose group of QXSTP,administered 1 time a day,continuous administration for 28 days.30 minutes after administration on the 26th day,except for the blank control group,sodium urate solution was injected into the rat ankle cavity to establish the gouty arthritis model of rats.The ankle swelling degree,gait score,serum urea(UA),TNF-αand IL-6 contents and histopathological examination were performed respectively.12 hours and 24 hours after modeling,compared with the model control group,the swelling degree of ankle in rats with low,medium and high dose group of QXSTP decreased to varying degrees.After 24 hours,gait score of high/medium doses decreased significantly.The content of UA and TNF-αdecreased to varying degrees in high/medium doses,and the content of IL-6 in high dose was significantly reduced.Compared with the model control group,high,medium and low doses preventive administration significantly reduced HE staining pathology scores.Under the experimental conditions,QXSTP has a therapeutic effect on gouty arthritis in rats caused by sodium urate.
