REV感染的SPF鸡免疫器官中免疫相关因子动态变化的研究

摘要 先天性免疫反应是机体抵御病原微生物感染的第一道防线。为了解禽网状内皮组织增生症病毒(REV)感染的SPF鸡早期的天然免疫反应,本研究利用qRT-PCR技术检测了REV感染的SPF鸡主要免疫器官中的免疫应答情况。结果显示,与病毒未感染组相比,感染鸡免疫器官中天然免疫受体TLR3和TLR7和Ⅱ型干扰素IFN-γ以及Viperin在病毒大量复制时,表达量显著升高;在病毒感染后第14天,胸腺、脾脏和法氏囊中炎性因子IL-6和IL-1β、抑炎因子IL-10及主要组织相容性复合物MHCⅠ的表达同时显著上调;在脾脏和法氏囊中第28天Ⅰ型干扰素IFN-α和IFN-β表达明显上升。此外,还发现,REV感染鸡的脾脏在感染后1~3 d内,TLR7、IL-1β、IFN-γ、MHCⅠ和MHCⅡ分子都出现不同程度的抑制,后期随病毒含量增加出现明显上升。上述结果说明,宿主体内主要免疫器官中细胞因子的表达与病毒复制有着紧密的相关性。本研究结果为深入探索REV的致病机制提供了基础资料。 Innate immune system is the body’s first line of defense against infection by pathogenic microorganisms.In order to investigate the early innate immune response of SPF chickens infected with avian reticuloendotheliosis virus(REV),the q RT-PCR was used to detect the immune response in the main immune organs of SPF chickens infected with REV.In result,the expression levels of the receptors TLR3 and TLR7,IFN-γ and viperin in the immune organs of infected chickens were significantly higher than those in the uninfected group.On day 14 after infection,the expression of inflammatory factors IL-6 and IL-1β,anti-inflammatory factor IL-10 and major histocompatibility complex MHCⅠin thymus,spleen and bursa of fabricius were significantly up-regulated simultaneously.In spleen and bursa on day 28 post-infection,the expression of IFN-α and IFN-β increased significantly.In addition,we also found that the spleen of chickens infected with REV showed different degrees of gene inhibition of TLR7,IL-1β,IFN-γ,MHCⅠ and MHCⅡ molecules within 1 to 3 days,but increased significantly with the increase of virus content in the later infection period.All the experimental results indicated that the expression of cytokines in the main immune organs of the host is closely related to the replication of the virus.The results of this study provide reference data for further study on the pathogenic mechanism of REV.
