摘要
目的:探讨原发性肝癌患者高尔基体蛋白73(GP73)的表达与外周血淋巴细胞亚群的相关性及其可能的临床意义。方法:收集2018年6月~2020年7月确诊为原发性肝癌患者129例及健康对照者36例,检测外周血血清GP73的表达量并根据临界值将原发性肝癌患者划分为阳性组和阴性组;流式细胞术检测外周血淋巴细胞亚群百分数。采用SPSS17.0统计软件进行数据分析。结果:阳性组、阴性组与健康对照组GP73的表达量分别为(254.773±128.566)mg/ml、(65.591±26.158)mg/ml与(28.137±19.115)mg/ml,阳性组显著高于阴性组与对照组(P<0.001);对照组与阴性组差异无统计学意义(P=0.103);阳性组CD3^(+)T淋巴细胞百分数显著低于对照组(P=0.010),与阴性组差异无统计学意义(P=0.298)。阳性组NKT淋巴细胞百分数显著高于阴性组(P=0.015),与对照组差异无统计学意义(P=0.220)。CD19^(+)B细胞、NK细胞、CD4^(+)T细胞、CD8^(+)T细胞百分数组间差异均无统计学意义。结论:GP73阳性的原发性肝癌患者CD3^(+)T淋巴细胞百分数减少,临床表现为免疫抑制状态;肝癌患者NKT淋巴细胞百分数与健康人群无差异,但GP73阳性的肝癌患者NKT淋巴细胞显著高于GP73阴性原发性肝癌患者,可能暗示GP73在原发性肝癌中的表达与NKT淋巴细胞的免疫调控有关。
Objective:To investigate the correlation between the expression of Golgi protein 73(GP73)and peripheral blood lymphocyte subsets in patients with primary hepatocellular carcinoma(PHC)and its possible clinical significance.Methods:Total of 129 patients diagnosed with PHC were selected as research group while 36 healthy follow-up individuals were randomly selected as control group from June 2018 to July 2020.The expression of serum GP73 was detected by enzymatic immunoassay and the PHC group was divided into positive and negative groups according to the critical value.The percentage of peripheral blood lymphocyte subsets was detected by flow cytometry.SPSS17.0 statistical software was used for data analysis.Results:The expression level of GP73 in the positive group(254.773±128.566)mg/mL was significantly higher than that in the negative group(65.591±26.158)mg/mL and healthy control group(28.137±19.115)mg/mL(P<0.001).There was no significant difference between the control group and the negative group(P=0.103).The percentage of CD3^(+)T lymphocytes in the positive group was significantly lower than that in the control group(P=0.010),but there was no significant difference between the positive group and the negative group(P=0.298).The percentage of NKT lymphocytes in the positive group was significantly higher than that in the negative group(P=0.015),but there was no significant difference between the positive group and the healthy control group(P=0.220).There was no significant difference in the percentages of CD19^(+)B lymphocytes,NK lymphocytes,CD4^(+)T lymphocytes,and CD8^(+)T lymphocytes among groups.Conclusion:Patients with GP73 positive expression in PHC showed decreased percentage of CD3^(+)T lymphocytes which is related to immunosuppression.There was no difference in the percentage of NKT lymphocytes in PHC patients and healthy people,but the percentage of NKT lymphocytes in patients of GP73-positive expression was significantly higher than those in patients of GP73-negative expression.The expression of GP73 in PHC may be related to the immune regulation of NKT lymphocytes.
