摘要
以奶牛乳腺上皮细胞(Dairy cow mammary epithelial cells,DCMECs)为研究模型,探讨microRNA-148b对DCMECs增殖和β-酪蛋白合成的影响。应用脂质体转染技术将miR-148b转染DCMECs,qRT-PCR观测其表达量,采用双荧光素酶实验验证miR-148b与转化生长因子B2(TGFB2)基因的靶向关系,分别应用EdU实验、酶联免疫吸附实验检测DCMECs的增殖以及β-酪蛋白含量。双荧光素酶实验结果证明TGFB2为miR-148b的靶基因,EdU实验结果显示,miR-148b可以促进DCMECs的增殖,ELISA结果表明过表达miR-148b可以促进β-酪蛋白合成。
Using Dairy cow mammary epithelial cells(DCMECs)as a research model to explore the effects of microRNA-148b on the proliferation andβ-casein synthesis in DCMECs.Liposomal transfection technology was used to transfect miR-148b into DCMECs,qRTPCR was used to test expression of miR-148b in DCMECs.The dual luciferase experiment verified the relationship between miR-148b and TGFB2 gene.The proliferation andβ-casein content of DCMECs were detected by EdU and enzyme-linked immunosorbent assay.The result of dual luciferase experiment showed that miR-148b has a targeting relationship with TGFB2 gene.The result of EdU experiment showed that miR-148b can promote the proliferation of DCMECs,and the result of ELISA assay showed that overexpression of miR-148b can promoteβ-casein synthesis.
