胶红酵母CYJ03的体外体内抗氧化活性研究 被引量:1

摘要 旨在深入研究海洋源胶红酵母CYJ03的抗氧化能力,挖掘其开发成抗氧化产品的潜力。通过测定CYJ03的过氧化氢耐受性,及其发酵上清液、完整细胞和色素提取物3种不同组分的还原力、自由基清除能力和Fe^(2+)螯合能力来评价该菌株的体外抗氧化能力;通过在罗非鱼日粮中添加CYJ03,测定罗非鱼的生长性能以及血清和肝脏的总抗氧化能力(T-AOC)、抗氧化酶活性来评价该菌株的体内抗氧化能力。CYJ03具有极强的过氧化氢耐受性,当H_(2)O_(2)浓度为13 mmol/L时仍有2.58%的存活率;在其3种组分中,发酵上清液具有最强的还原力、二苯代苦味肼基自由基(DPPH·)清除力和Fe^(2+)螯合能力,分别达到0.26±0.02、(92.92±1.03)%和(83.81±0.73)%;色素提取物具有最强的·OH清除力(44.58±0.98)%。在日粮中添加0.5%、1%和2%的CYJ03饲喂红罗非鱼60 d,能够显著提高罗非鱼的增重率、特定生长率和肝体比,显著降低饲料系数(P<0.05);同时能够显著提高罗非鱼血清和肝脏的T-AOC以及过氧化氢酶(CAT)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)的活性,且以1%添加量最为显著。胶红酵母CYJ03的各组分均具有体外抗氧化活性,并且菌体可有效提高罗非鱼的生长性能、提高T-AOC和抗氧化酶活性,具有良好的体内抗氧化作用,具有潜在开发成微生物产品的价值。 The purpose of this study was to deeply investigate the antioxidative effects of Rhodotorula mucilaginosa CYJ03 and to tap its potential to develop antioxidative products.The antioxidant activities in vitro of CYJ03 were evaluated by measuring the tolerance to hydrogen peroxide,the reducing powers,the abilities for scavenging free radicals,Fe^(2+) chelating capacity of its fermentation supernatant,intact cells and pigment extract,respectively.Whereas the antioxidant activities in vivo were evaluated through measuring the growth performance,total antioxidant capacity(T-AOC)and antioxidant enzymes activities in serum and liver after the supplemental test of tilapia with R.mucilaginosa CYJ03 addition to the diet.The results indicated that R.mucilaginosa CYJ03 had strong hydrogen peroxide tolerance with the survival rate maintained at 2.58%,when it was challenged by H_(2)O_(2) at concentration up to 13 mmol/L.The fermentation supernatant showed the strongest abilities of reducing power,DPPH·scavenging powers and Fe^(2+) chelating capacity,exemplified by the performances as to 0.26±0.02,(92.92±1.03)%and(83.81±0.73)%respectively.Whereas the pigment extract had delivered the strongest scavenging power of·OH(44.58±0.98)%.Feeding tilapia with 0.5%,1%and 2%strain CYJ03 for 60 days could significantly increased the growth ratio,specific growth rate and hepatosomatic index of tilapia,and significantly reduced the feed conversion ratio(P<0.05).At the same time,the supplement of CYJ03 significantly improved the activities of T-AOC,catalase(CAT),superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)in serum and liver of tilapia,respectively,And the addition of 1%was the most significant.Each component of R.mucilaginosa CYJ03 had antioxidant activity in vitro,and the preparations derived from CYJ03 could effectively improve the growth performance of tilapia,increase the T-AOC and antioxidant enzymes activities,had good antioxidantive effects in vivo.The results of this work had the potential application value of CYJ03 to develop into microbial products.
