急性肺栓塞大鼠肺组织中H-FABP表达的变化及意义 被引量:2

摘要 目的研究急性肺栓塞(APE)肺组织中心型脂肪酸结合蛋白(H-FABP)表达的变化及意义。方法选择成年雄性SD大鼠,随机分为对照组、APE组、NC-shRNA组、NC-shRNA+APE组、H-FABP-shRNA+APE组,采用自体血栓法建立APE模型,给予shRNA慢病毒尾静脉注射。检测血清血小板活化指标P-选择素(CD62P)、溶酶体颗粒糖蛋白63(CD63)、血栓素A2(TXA2),肺组织病理改变及心型脂肪酸结合蛋白(H-FABP)、核因子-κB(NF-κB)的表达水平,单核细胞趋化蛋白-1(MCP-1)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)的含量。结果 APE组大鼠肺组织中H-FABP的表达水平高于对照组(P<0.05),尾静脉注射H-FABP-shRNA腺病毒有效敲低APE大鼠肺组织中H-FABP的表达;NC-shRNA+APE组大鼠肺组织出现典型病理改变,血清CD62P、CD63、TXA2的含量及肺组织中H-FABP、NF-κB的表达水平、MCP-1、TNF-α、IL-1β的含量显著高于NC-shRNA组(均P<0.05);H-FABP-shRNA+APE组大鼠肺组织的病理改变改善,血清CD62P、CD63、TXA2的含量及肺组织中H-FABP、NF-κB的表达水平、MCP-1、TNF-α、IL-1β的含量显著低于NC-shRNA+APE组(均P<0.05)。结论 APE大鼠肺组织中H-FABP高表达与发病过程中血小板活化、炎症反应激活有关。 Objective To study the expression of heart-type fatty acid binding protein(H-FABP)in lung tissue of acute pulmonary embolism(APE)and its significance.Methods Adult male SD rats were randomly divided into control group, APE group, NC shRNA group, NC shRNA+APE group and H-FABP-shRNA+APE group.APE model was established by autologous thrombus method and shRNA lentivirus was injected into tail vein.The levels of CD62 P,CD63,TXA2 in serum, the pathologic change of lung, the expression of H-FABP,NF-κB and the contents of MCP-1,TNF-α and IL-1β in lung tissue were detected.Results The expression levels of H-FABP in lung tissue of APE rats were higher than that of control group(P<0.05).The lung tissue of NC-shRNA+APE group showed typical pathological changes of lung, the levels of CD62 P,CD63 and TXA2 in serum, the expression levels of H-FABP and NF-κB,the levels of MCP-1,TNF-α and IL-1β in lung tissue were significantly higher in NC-shRNA+APE group than in NC-shRNA group(P<0.05).The pathological changes of lung tissue in H-FABP-shRNA+APE group were less severe.The contents of CD62 P,CD63 and TXA2 in serum, the expression levels of H-FABP and NF-κB and the levels of MCP-1,TNF-α and IL-1β in lung tissue were significantly lower in the H-FABP-shRNA+APE group than in the NC-shRNA+APE group(P<0.05).Conclusion The high expression of H-FABP in lung tissue of APE rats is related to the activation of platelet and inflammatory reaction.
