胃癌组织lncRNA MAFG-AS1、miR-143-3p表达变化及其临床意义

摘要 目的观察胃癌组织长链非编码RNA(lncRNA)碱性亮氨酸拉链家族转录因子V-Maf鸟类肌筋膜纤维肉瘤癌基因同源物G-反义核糖核酸1(MAFG-AS1)、微小RNA-143-3p(miR-143-3p)表达变化,并探讨二者表达与患者临床病理特征和预后的关系。方法选择胃癌患者105例,取手术切除的胃癌组织及其配对的癌旁正常组织,采用RT-qPCR技术检测lncRNA MAFG-AS1、miR-143-3p表达。收集胃癌患者临床病理资料和术后随访资料,分析胃癌组织lncRNA MAFG-AS1、miR-143-3p表达与患者临床病理特征和预后的关系。结果胃癌组织lncRNA MAFG-AS1相对表达量高于癌旁正常组织,miR-143-3p相对表达量低于癌旁正常组织(P均<0.05)。胃癌组织lncRNA MAFG-AS1、miR-143-3p表达与肿瘤组织分化程度、浸润深度、pTNM分期、淋巴结转移有关(P均<0.05),而与性别、年龄、肿瘤直径无关(P均>0.05)。胃癌组织lncRNA MAFG-AS1表达与miR-143-3p表达呈负相关关系(r=-0.699,P<0.05)。以lncRNA MAFG-AS1、miR-143-3p相对表达量的均数为临界值,将患者分为lncRNA MAFG-AS1高表达者55例与lncRNA MAFG-AS1低表达者50例、miR-143-3p高表达者48例与miR-143-3p低表达者57例。lncRNA MAFG-AS1高表达者3年总生存率低于lncRNA MAFG-AS1低表达者,miR-143-3p高表达者3年总生存率高于miR-143-3p低表达者(χ^(2)分别为4.783、5.383,P均<0.05)。结论胃癌组织lncRNA MAFG-AS1表达上调、miR-143-3p表达下调,二者表达变化与胃癌恶性生物学行为和患者预后不良有关。 Objective To investigate the expression of long non-coding RNA( lncRNA) MAF b ZIP transcription factor G antisense RNA 1( MAFG-AS1) and microRNA-143-3 p( miR-143-3 p) in the gastric cancer,and to explore the relationship between their expression and clinicopathological features and prognosis. Methods Totally 105 patients with gastric cancer were selected,and the surgically resected gastric cancer tissues and their matched adjacent normal tissues were taken. The expression levels of lncRNA MAFG-AS1 and miR-143-3 p were detected by RT-qPCR. The clinicopathological data and postoperative follow-up data of patients with gastric cancer were collected,and the relationships between the expression levels of lncRNA MAFG-AS1 and miR-143-3 p in gastric cancer and clinicopathological features and prognosis were analyzed. Results The relative expression of lncRNA MAFG-AS1 in the gastric cancer tissues was higher than that in the adjacent normal tissues,and the relative expression of miR-143-3 p was lower than that in the adjacent normal tissues( both P < 0. 05). The expression of lncRNA MAFG-AS1 and miR-143-3 p in gastric cancer was related to the degree of differentiation,depth of invasion,p TNM stage and lymph node metastasis( all P < 0. 05),but not to gender,age or tumor diameter( all P > 0. 05). There was a negative correlation between lncRNA MAFG-AS1 expression and miR-143-3 p expression in gastric cancer( r =-0. 699,P < 0. 05). Taking the average of the relative expression of lncRNA MAFGAS1 and miR-143-3 p as the critical value,the patients were divided into 55 cases with high expression of lncRNA MAFGAS1,50 cases with low expression of lncRNA MAFG-AS1,48 cases with high expression of miR-143-3 p,and 57 cases with low expression of miR-143-3 p. The 3-year overall survival rate of those with high expression of lncRNA MAFG-AS1 was lower than that of those with low expression of lncRNA MAFG-AS1,and the 3-year survival rate of those with high expression of miR-143-3 p was higher than that of those with low expression of miR-143-3 p( χ^(2)= 4. 783 and 5. 383,respectively;both P < 0. 05). Conclusions The expression of lncRNA MAFG-AS1 was up-regulated and miR-143-3 p was down-regulated in gastric cancer. Their expression changes were related to the malignant biological behavior of gastric cancer and poor prognosis of patients.
