基于网络药理学探讨蛇莓-白英治疗银屑病的作用机制 被引量:1

Mechanism of Duchesnea Indica-Solanum Lyratum for the Treatment of Psoriasis Based on Network Pharmacology

摘要 目的探讨“蛇莓-白英”药对治疗银屑病的作用机制。方法文本挖掘方式检索中药分子作用机制网络药理学在线分析平台(Batman-TCM)数据库,收集蛇莓所含有的有效成分及靶点基因,通过设置Score cutoff≥10和P≥0.05为条件进行筛选。在中国知网上进行文献检索,找出白英的主要化学成分。通过化学成分靶点预测和注释,利用Cytoscape3.7.1软件进行靶点-疾病网络的构建及分析,利用STRING在线分析平台进行蛋白质-蛋白质相互作用网络(PPI)网络构建,预测重要的成分及靶点。利用bioconductor程序包中的clusterProfiler包对蛇莓-白英药对的靶点进行基因本体(GO)功能富集分析和基于京都基因与基因组百科全书(KEGG)通路富集分析。结果筛选出蛇莓-白英药对17个有效成分(蛇莓6个、白英11个)对应靶点164个,其中与银屑病相关靶点101个,富集分析可得相关的20个潜在信号通路包括有机酸结合、细胞因子活性、细胞因子受体结合、表皮生长因子酪氨酸激酶抑制剂抵抗通路(EGFR-TKIs)、白细胞介素-17(IL-17)信号通路、血管内皮生长因子(VEGF)信号通路等。结论蛇莓-白英药对治疗银屑病与多靶点、多通路相关,这为深入探讨其作用的物质基础以及作用机制提供了参考。 Objective In order to explore the mechanism of"Duchesnea indica-Solanum lyratum"medicine in treating psoriasis.Methods Text mining was used to retrieve the Batman-TCM database to collect the active ingredients and target genes contained in Duchesnea indica.By setting score cutoff greater or equal to 10 and screening was performed with P value greater or equal to 0.05.A literature search was conducted on China Knowledge Network to find the main chemical constituents of Solanum lyratum.Through chemical component target prediction and annotation,Cytoscape 3.7.1 software was used to construct and analyze the target-disease network,and the STRING online analysis platform was used to construct the PPI network to predict important components and targets.The clusterProfiler package in the bioconductor package was used to perform gene ontology(GO)function enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis on targets of the Duchesnea indica-Solanum lyratum drug pair.Results Seventeen active components of Duchesnea indica-Solanum lyratum medicine were screened out(6 Duchesnea indica and 11 Solanum lyratum),corresponding to 101 of 164 targets related to psoriasis,and 20 related by enrichment analysis potential signaling pathways include organic acid synthesis,cytokine activity,cytokine receptor binding,epidermal growth factor tyrosine kinase inhibitor resistance,interleukin(IL)-17 signaling pathway,and vascular endothelial growth factor(VEGF)signaling pathway.Conclusion It is revealed that Duchesnea indica-Solanum lyratum medicine is related to multiple targets and multiple pathways in the treatment of psoriasis,which provide references for further discussion of its material basis and mechanism.
