短暂缺氧对3日龄SD大鼠海马胰岛素受体和β-catenin及NeuroD1表达的影响

摘要 目的:探讨短暂缺氧对3日龄SD大鼠海马胰岛素受体和β-catenin及NeuroD1表达的影响。方法:将3日龄SD大鼠置于8%O2+92%N2混合气体中2 h,建立缺氧性脑损伤模型(模型组),对照组为将3日龄SD大鼠置于缺氧箱2 h,不通入混合气体。2组大鼠分别于缺氧后2、4、24h断头取脑,采用免疫印迹和免疫荧光法检测海马active-β-catenin、NeuroD1、胰岛素受体蛋白表达的变化。结果:免疫印迹结果显示,缺氧后2 h,模型组与对照组active-β-catenin、NeuroD1、胰岛素受体蛋白表达量差异无统计学意义;缺氧后4h,active-β-catenin和NeuroD1蛋白表达量增高,胰岛素受体蛋白表达量差异无统计学意义;缺氧后24h,active-β-catenin、NeuroD1和胰岛素受体蛋白表达量均增高。结论:短暂缺氧能刺激active-β-catenin、NeuroD1和胰岛素受体蛋白表达;胰岛素受体表达量应激性增高可能受Wnt/β-catenin/NeuroD1通路的影响。 Objective:To investigate the effects of transient hypoxia on the expression of insulin receptor,β-catenin and NeuroD1 in hippocampus of 3-day-old SD rats.Methods:Three-day-old SD rats were exposed to 8%O;+92%N;mixture for 2 hours to establish hypoxic brain injury model.Rats at the same age placed in the hypoxic tank for 2 h without the mixed gas was as control.The expression of active-β-catenin,NeuroD1 and insulin receptor in hippocampus were detected by Western blotting and immunofluorescence.Results:Western blotting analysis showed that there was no significant difference in the expression of three above mentioned proteins among the three groups at 2 h after hypoxia.At 4 h after hypoxia,the expression of active-β-catenin and NeuroD1 increased,but the expression of insulin receptor had no significant difference.At 24 h after hypoxia,the expression of active-β-catenin,NeuroD1 and insulin receptor increased.Conclusion:Transient hypoxia can stimulate the expression of active-β-catenin,NeuroD1 and insulin receptor,which may be affected by Wnt/β-catenin/NeuroD1 pathway.
