非小细胞肺癌合并肺部感染患者组织中血管紧张素转换酶2的表达及机制 被引量:3

Expression and mechanism of angiotensin converting enzyme 2 in non-small cell lung cancer patients with pulmonary infection

摘要 目的:探讨血管紧张素转换酶2(ACE2)在非小细胞肺癌(NSCLC)合并肺部感染的表达及作用。方法:2017年12月至2019年12月来宁夏医科大学总医院就诊的NSCLC患者纳入此次研究。患者分为NSCLC组(病理诊断为NSCLC,临床和影像学诊断不合并肺部感染)和合并感染组(病理诊断为NSCLC,临床和影像学诊断合并肺部感染);免疫组织化学检测ACE-2的表达;流式细胞术检测红系前体细胞、CD45^(+)红系前体细胞、T细胞、CD3^(+)T细胞、CD8^(+)T细胞比例以及活性氧(ROS)的含量。结果:免疫组织化学结果显示NSCLC组ACE2平均光密度为14.68,合并感染组为131.87;流式细胞术结果显示NSCLC组CD45^(+)红系前体细胞比例为4.86%,合并感染组为9.67%;相关性分析的结果显示NSCLC组织内ACE2的表达与外周血CD45^(+)红系前体细胞比例呈正相关;NSCLC组CD3^(+)T细胞和CD8^(+)T细胞比例分别为60.98%和34.52%,合并感染组比例分别为49.87%和20.45%;加入ROS抑制剂后CD3^(+)T细胞和CD8^(+)T细胞比例分别为64.33%和41.75%。相对于合并感染组,差异均具有统计学意义。结论:ACE2在合并肺部感染的NSCLC组织内高表达,其可通过CD45^(+)红系前体细胞和ROS,降低CD3^(+)T细胞和CD8^(+)T细胞比例。 Objective:To investigate the expression and role of angiotensin converting enzyme 2(ACE2)in non-small cell lung cancer(NSCLC)with pulmonary infection.Methods:NSCLC patients who came to our hospital from December 2017 to December 2019 were included in this study.Patients were divided into the NSCLC group(pathological diagnosis was NSCLC,clinical and imaging diagnosis was not combined with pulmonary infection)and the combined infection group(pathological diagnosis was NSCLC,clinical and imaging diagnosis was combined with pulmonary infection).Immunohistochemistry was used to detect the expression of ACE-2.Flow cytometry was used to detect the proportion of erythroid precursor cells,CD45^(+)erythroid precursor cells,CD3^(+)T cells,CD8^(+)T cells and the content of ROS.Results:The average optical density of ACE2 was 14.68 in NSCLC group and 131.87 in the combined infection group.The results of flow cytometry showed that the proportion of CD45^(+)erythroid precursor cells was 4.86%in NSCLC group and 9.67%in the combined infection group.The results of correlation analysis showed that the expression of ACE2 in NSCLC tissues was positively correlated with the proportion of peripheral blood CD45^(+)erythroid precursor cells.The proportion of CD3^(+)T cells and CD8^(+)T cells was 60.98%and 34.52%in NSCLC group,49.87%and 20.45%in the combined infection group,respectively.The proportion of CD3^(+)T cells and CD8^(+)T cells treated with the ROS inhibition was 64.33%and 41.75%,respectively.Compared with the combined infection group,the differences were statistically significant.Conclusion:ACE2 is highly expressed in NSCLC tissues with pulmonary infection.It can reduce the proportion of CD3^(+)T cells and CD8^(+)T cells through CD45^(+)erythroid precursor cells and ROS.
