摘要
目的:探讨异甘草酸镁(MgIG)对四氯化碳(CCl;)致大鼠肝纤维化模型肝细胞数量与体积的影响。方法:雄性SD大鼠随机分为对照组、模型组、治疗组,治疗组给予CCl4皮下注射,同时每日腹腔注射MgIG 30 mg/kg。各组大鼠在造模5周后取材,从每只大鼠等距抽选4个2 mm厚的肝组织块制作石蜡包埋切片,行Masson染色,采用体视学方法测量大鼠肝细胞体积与数量,以及肝细胞胞质内脂滴体积。结果:与对照组比较,模型组、治疗组大鼠肝细胞的平均体积分别显著增加了91.9%和105.0%,肝细胞胞质内脂滴的总体积分别显著增加了5.27倍与4.83倍,肝细胞的总数量分别显著减少了40.0%和42.4%;与模型组比较,治疗组大鼠每个肝细胞平均体积、肝细胞胞质内脂滴的总体积以及肝细胞的总数量之间的差异均无统计学意义。结论:CCl4所致大鼠肝纤维化伴随着肝细胞肿胀、脂肪变性及肝细胞数量减少等病理改变,MgIG对CCl4致肝细胞的病理改变无显著作用。
Objective:To investigate the effect of intervention of magnesium isoglycyrrhizinate(MgIG)on the number and volume of hepatocytes in carbon tetrachloride(CCl4)induced hepatic fibrosis model in rats.Methods:Male SD rats were randomly divided into a control group(5 rats),a model group(6 rats)and a treatment group(5 rats,injected 30 mg/kg MgIG every day at the start of building models with CCl;).Five weeks after the initial injection,four liver tissue blocks were sampled in a systematic(equally spaced)manner and were embedded in paraffin.Then,the paraffin-embedded sections were prepared and stained with Masson trichrome staining.The volume and quantity of hepatocytes and the volume of lipid droplets in the cytoplasm of hepatocytes were measured by stereological method.Results:Compared with the control group,the average volume of each hepatocyte in the model group and MgIG treatment group increased by 91.9%and 105.0%respectively,the total volume of lipid droplets in the cytoplasm of hepatocytes increased by 5.27 and 4.83 times respectively.Compared with the model group,the average volume of each hepatocyte and the total volume of lipid droplets in the cytoplasm of hepatocytes in the treatment group had no significant changes.Compared with the control group,the total number of liver cells in the model group and the treatment group were significantly reduced by 40.0%and 42.4%,respectively.Compared with the model group,there were no significant differences in the average volume of each hepatocyte,the total volume of lipid droplets and the total number of hepatocytes in the treatment group.Conclusion:CCl;induced liver fibrosis in rats is accompanied by pathological changes such as hepatocyte swelling,steatosis,and decrease in the number of liver cells.MgIG has no significant effect on the CCl;induced pathological changes of liver cells.
