摘要
目的探讨帕金森病(PD)患者血清miR-103a-3p、miR-486-5p水平变化及临床意义。方法选取183例PD患者为PD组,根据Hoehn-Yahr分级分为重度组(n=53)、中度组(n=71)、轻度组(n=59),另选取75例体检健康者为对照组。采用qRT-PCR检测血清miR-103a-3p、miR-486-5p,采用Spearman相关分析PD患者血清miR-103a-3p、miR-486-5p水平与Hoehn-Yahr分级的相关性,采用受试者工作特征(ROC)曲线分析血清miR-103a-3p、miR-486-5p水平对PD的诊断价值。结果与对照组比较,PD组血清miR-103a-3p、miR-486-5p水平降低(P均<0.05)。轻度组、中度组、重度组血清miR-103a-3p、miR-486-5p水平依次降低(P均<0.001)。PD患者血清miR-103a-3p、miR-486-5p水平与Hoehn-Yahr分级呈负相关(r s分别为-0.865、-0.815,P均<0.001)。miR-103a-3p+miR-486-5p、miR-103a-3p、miR-486-5p诊断PD的曲线下面积分别为0.868、0.799、0.775,miR-103a-3p+miR-486-5p诊断PD的曲线下面积大于miR-103a-3p、miR-486-5p单独诊断(P均<0.001)。结论miR-103a-3p、miR-486-5p在PD患者血清中水平低,与PD病情进展密切相关,可作为PD诊断指标。
Objective To investigate the expression changes of serum miR-103a-3p and miR-486-5p in patients with Parkinson′s disease(PD)and and their clinical significance.Methods Totally 183 PD patients were selected as the PD group and then were divided into the severe(n=53),moderate(n=71),and mild(n=59)groups according to Hoehn-Yahr classification,and 75 other physically healthy individuals were selected as the control group.The qRT-PCR was used to detect serum miR-103a-3p and miR-486-5p expression.Spearman′s correlation coefficient was used to analyze the correlation between serum miR-103a-3p,miR-486-5p expression and Hoehn-Yahr classification in PD patients.Receiver operating characteristic(ROC)curve was used to analyze the diagnostic value of serum miR-103a-3p and miR-486-5p expression for PD.Results Compared with the control group,serum miR-103a-3p and miR-486-5p expression decreased in the PD group(both P<0.05).Serum miR-103a-3p and miR-486-5p expression decreased gradually in the mild,moderate and severe groups(all P<0.001).Serum miR-103a-3p and miR-486-5p expression levels in PD patients were negatively correlated with Hoehn-Yahr classification(r s=-0.865,-0.815;all P<0.001).The area under the curve of miR-103a-3p,miR-486-5p,and miR-103a-3p+miR-486-5p in diagnosis of PD was 0.799,0.775,and 0.868,respectively.The area under the curve of miR-103a-3p+miR-486-5p in diagnosis of PD was greater than that of miR-103a-3por miR-486-5p alone(both P<0.001).Conclusion Serum miR-103a-3p and miR-486-5p expression levels are low in PD patients,which are closely related to the progression of PD and can be used as diagnostic indicators of PD.
