摘要
目的探究miR-107对乳腺癌细胞侵袭及上皮-间质转化(EMT)的影响和机制。方法利用荧光定量PCR方法分析miR-107在正常乳腺细胞及乳腺癌细胞系中的表达差异;HS578T细胞分别转染对照miRNA和miR-107抑制剂,利用划痕实验分析细胞迁移能力,用Transwell实验分析细胞侵袭能力,用实时荧光定量PCR方法检测细胞EMT标志物的表达水平,用蛋白质免疫印迹实验检测细胞PTEN/AKT信号通路。结果与正常乳腺细胞相比,miR-107在乳腺癌细胞株中高表达;沉默miR-107的表达抑制HS578T细胞的迁移、侵袭及EMT,促进PTEN/AKT信号通路。结论miR-107在乳腺癌细胞中高表达,沉默miR-107表达通过PTEN/AKT信号通路抑制乳腺癌细胞迁移、侵袭和EMT。
Objective To explore the effect and mechanism of miR-107 on invasion and epithelial-mesenchymal transformation(EMT)of breast cancer cells.Methods Expression of miR-107 in normal breast cell and breast cancer cell lines were measured by qPCR;HS578T cells were transfected with control miRNA or miR-107 inhibitor,wound-healing assay was used to detect cell migration,and Transwell assay was used to measure cell invasion capability.The biomarkers of EMT progress were detected by qPCR,and the PTEN/AKT signal pathway were detected by immunoblotting assay.Results MiR-107 was highly expressed in breast cancer cell lines compared with normal breast cells.The migration,invasion and EMT of HS578T cells were inhibited by the silencing of Mir-107 expression,as well as the PTEN/AKT signaling pathway.Conclusions The expression of miR-107 is up-regulated in breast cancer cells.Silencing miR-107 suppress breast cancer cell migration,invasion and EMT through PTEN/AKT signals.
