神香苏合丸对心肌梗死大鼠肠道菌群的影响

Effect of Shenxiang Suhe Pills on intestinal flora in rats with myocardial infarction

[目的]探索神香苏合丸对心肌梗死大鼠肠道菌群的影响。[方法]运用16S扩增子测序技术检测神香苏合丸给药14 d后各组心肌梗死大鼠肠道菌群的变化,通过体外培养实验检测神香苏合丸对关键肠道微生物生长的影响。[结果]神香苏合丸对心肌梗死大鼠肠道菌群具有改善作用,主要表现为降低心肌梗死大鼠肠道中厚壁菌门丰度,增加拟杆菌门丰度,降低厚壁菌门与拟杆菌门比值;降低链球菌、罗姆布茨菌、毛螺菌科、单球菌属丰度(P<0.05);增加罗斯氏菌和乳球菌属丰度(P<0.05)。此外,乳杆菌属作为肠道中的优势菌群,神香苏合丸对其丰度具有上调作用。体外验证实验结果显示神香苏合丸可以提高约氏乳杆菌EU03、鼠乳杆菌EU04的生长率。[结论]神香苏合丸对心肌梗死大鼠肠道菌群有改善作用,尤其能够促进肠道益生菌的生长,推测肠道益生菌可能是神香苏合丸治疗心肌梗死的潜在靶标。

[Objective]Explore the effect of Shenxiang Suhe Pills on the intestinal flora of rats with myocardial infarction.[Methods]The 16S amplicon sequencing technology was used to detect the changes of the intestinal flora of the rats with myocardial infarction in each group 14 days after the administration of Shenxiang Suhe Pills,the influence of Shenxiang Suhe Pills on key intestinal microorganisms was detected by in vitro culture experiments.[Results]Shenxiang Suhe Pills can improve the intestinal flora of rats with myocardial infarction,mainly by reducing the abundance of Firmicutes in the intestine of rats with myocardial infarction,increasing the abundance of Bacteroidetes,and reducing the abundance of the ratio of Firmicutes/Bacteroides;reducing the abundance of Streptococcus,Romboutsia,Lachnospiraceae,and Monoglobus(P<0.05);significantly increasing the abundance of Roseburia and Lactococcus(P<0.05).In addition,Lactobacillus is the dominant flora in the intestine,and Shenxiang Suhe Pills can up-regulate its abundance.The results of in vitro experiments showed that Shenxiang Suhe Pills can increase the growth rate of Lactobacillus johnsonii EU03 and Lactobacillus murinus EU04.[Conclusion]Shenxiang Suhe Pills can improve the intestinal flora of rats with myocardial infarction,especially promoting the growth of intestinal probiotics.It is speculated that the intestinal probiotics may be the potential targets of Shenxiang Suhe Pills in the treatment of myocardial infarction.
