摘要
为了探究Hippo信号通路在猪卵泡发育中发挥的作用,利用荧光定量PCR技术(Fluorescence quantitative PCR,qPCR)对Hippo通路核心因子(LATS1∕2、MOB1、MST1∕2、SAV1和YAP)mRNA在大白猪各发育阶段卵泡中的表达进行检测。结果表明:Hippo信号通路的7个核心因子在不同直径猪卵泡中的表达均达到差异显著水平,其中LATS2随着卵泡的不断生长表达量逐渐增加,而MOB1、MST1、MST2以及YAP则随着卵泡的不断生长表达量逐渐下降。初步表明Hippo信号通路核心因子LATS2在猪卵泡发育中发挥正调控作用,而MOB1、MST1、MST2以及YAP则发挥着负调控作用。研究结果可为猪卵泡发育机制的解析提供一定的理论基础。
In order to explore the role of the Hippo signaling pathway in porcine follicle development,fluorescence quantitative PCR(qPCR)was used in this study to detect the mRNA expression of the core factors of Hippo pathway(LATS1∕2,MOB1,MST1∕2,SAV1 and YAP)in the follicular development of Large White Pigs.The results showed that the expression levels of the seven core factors of Hippo signaling pathway in different diameter pig follicles were significantly different,among which the expression level of LATS2 increased gradually with the growth of follicles,while the expression levels of MOB1,MST1,MST2 and YAP decreased gradually with the growth of follicles.The results preliminary indicated that LATS2,the core factor of Hippo signaling pathway,played a positive role in pig follicular development,while MOB1,MST1,MST2 and YAP played a negative role,which provided a theoretical basis for the analysis of pig follicular development mechanism.
