基于生物信息学方法探究ENO1基因在宫颈癌中的 表达及其作用机制

摘要 目的通过生物信息学方法分析α-烯醇化酶(ENO1)在宫颈癌中的表达及功能特性。方法利用基因表达谱数据动态分析(GEPIA)探究ENO1基因在宫颈癌及癌旁组织的表达,分析其与宫颈癌患者预后的关系,利用人蛋白质数据库(HPA)进一步分析ENO1蛋白在宫颈鳞癌和癌旁组织的表达情况,通过癌症基因组图谱(TCGA)数据库分析影响患者预后的因素,借助基因富集分析(GSEA)ENO1基因在宫颈癌发展过程中可能的调控通路,利用Linked Omics数据库探索其共表达基因。结果ENO1基因在宫颈癌组织中的表达水平高于癌旁组织,差异有统计学意义(P<0.05);ENO1基因高表达与患者的总生存期(OS)呈负相关(P<0.05);ENO1基因功能的发挥主要涉及烟酸和烟酰胺代谢、脂肪酸代谢、过氧化物酶体等一系列与代谢相关的生物学过程;共表达基因分析显示,ALDQA、NOC2L、TUBA1C等基因与ENO1基因表达呈正相关且自上而下依次排列,而RNF103、MCF2L、CAPN5等基因与该基因的表达呈负相关并自上而下依次排列。结论ENO1基因在宫颈癌中呈高表达,与宫颈癌的发生发展及预后存在一定关系,有望成为宫颈癌预后评估和治疗的潜在靶点。 Objective To analyze and explore the expression and functional characteristics ofα-enolase(ENO1)in cervical cancer through bioinformatics methods.Methods The gene expression profiling interactive analysis(GEPIA)was used to explore the expression of ENO1 gene in cervical cancer and adjacent tissues,and its relationship with the prognosis of cervical cancer patients was analyzed.The expression of ENO1 protein in cervical squamous cell carcinoma and adjacent tissues was further analyzed by human protein atlas(HPA).The factors affecting the prognosis of patients were analyzed by the cancer genome atlas(TCGA)database.The possible regulatory pathways of ENO1 gene in the development of cervical cancer were analyzed by gene set enrichment analysis(GSEA),and the co-expression genes were explored by Linked Omics database.Results The expression level of ENO1 gene in cervical cancer tissues was higher than that in adjacent tissues,and the difference was statistically significant(P<0.05).The high expression of ENO1 gene was negatively correlated with the overall survival(OS)of patients(P<0.05).The function of ENO1 gene mainly involved a series of metabolism-related biological processes such as niacin and nicotinamide metabolism,fatty acid metabolism,and peroxisome.Co-expression gene analysis showed that ALDQA,NOC2L,TUBA1C and other genes were positively correlated with the expression of ENO1 gene and ranked from top to bottom,while RNF103,MCF2L,CAPN5 and other genes were negatively correlated with the expression of this gene and ranked from top to bottom.Conclusion ENO1 gene is highly expressed in cervical cancer,which is related to the occurrence,development and prognosis of cervical cancer,and is expected to become a potential target for prognosis evaluation and treatment of cervical cancer.
