辛酰水杨酸纳米胶束制剂的制备及其含量测定 被引量:3

摘要 目的制备辛酰水杨酸纳米胶束制剂,测定胶束粒径并建立辛酰水杨酸的含量测定方法。方法在水溶液中制备自组装辛酰水杨酸纳米胶束,采用激光粒度仪测定其平均粒径和粒径分布,通过SEM察纳米胶束形态,采用HPLC法测定纳米胶束制剂中辛酰水杨酸的含量。结果辛酰水杨酸胶束平均粒径7.96 nm,粒径分布2~12 nm,SEM照片显示纳米胶束粒径较均匀;辛酰水杨酸含量测定HPLC法:Waters XBridge C_(18)(4.6 mm×150 mm,5μm)色谱柱;流动相:甲醇-乙腈-0.1 mol/L磷酸溶液(48:30:22);检测波长:225 nm;柱温:40℃;流速:1.0 ml/min;进样量:10μl。在4.1~40.1μg/ml范围内线性关系良好;平均回收率为100.49%,RSD为1.82%。结论自组装辛酰水杨酸纳米胶束粒径分布均匀,HPLC法测定纳米胶束制剂中辛酰水杨酸的含量,方法简便、快速、重复性好、准确度高。 Objective To prepare capryloyl salicylic acid(CSA)nanomicelles,measure the particle size of nanomicelles and establish an HPLC method for the determination of CSA in nanomicelles.Methods The self-assembled CSA nanomicelles was prepared in aqueous solution.The average particle size and particle size distribution were measured by Zetasizer.The morphology of nanomicelles was observed by SEM.The content of CSA in nanomicelles was determined by HPLC.Results The average particle size of CAS Nanomicelles was 7.96 nm and the particle size distribution was 2-12 nm.SEM photos showed that the particle size of nanomicelles was uniform.The HPLC method was carried out on a Waters XBridge C_(18) column(4.6 mm×150 mm,5μm).The mobile phase was methanol-acetonitrile-0.1 mol/L H_(3)PO_(4) solution(48:30:22)with the detection wavelength set at 225 nm,the column temperature was 40℃and the flow rate was 1.0 ml/min.The injection volume was 10μl.The linear range of CSA was 4.1-40.1μg/ml.The average recovery of CSA was 100.49%with RSD of 1.82%.Conclusion The self-assembled CSA Nanomicelles has uniform particle size distribution.The HPLC method for CSA determination is simple,rapid,reproducible and accurate.
