摘要
为了研究宫内发育迟缓(IUGR)仔猪小肠形态和屏障功能相关基因的表达特征,选取24窝"长白×大白"杂交新生仔猪,每窝选取1头IUGR仔猪和1头正常出生体重(NBW)仔猪,分别于7、21和28日龄屠宰8头IUGR仔猪和8头NBW仔猪,采集小肠样品进行分析。结果表明:1)与NBW仔猪相比,IUGR仔猪7日龄时空肠绒毛高度、21日龄时空肠和回肠绒毛高度显著降低(P<0.05),28日龄时空肠隐窝深度显著提高(P<0.05)。2)与NBW仔猪相比,IUGR仔猪7日龄时空肠黏膜闭锁小带蛋白-1(ZO-1)、封闭蛋白-1(claudin-1)、闭合蛋白(occludin)、白细胞介素-4(IL-4)和白细胞介素-8以及21日龄时空肠黏膜ZO-1和occludin、回肠黏膜ZO-1的mRNA相对表达量显著下调(P<0.05);28日龄时空肠黏膜肿瘤坏死因子-α和回肠黏膜白细胞介素-1β的mRNA相对表达量显著上调(P<0.05),回肠黏膜IL-4的mRNA相对表达量显著下调(P<0.05)。综上所述,IUGR仔猪的空肠和回肠形态受损,紧密连接蛋白基因表达下调,免疫细胞因子表达异常,进而影响小肠屏障功能。
The present study was conducted to determine the profiles of small intestinal morphology and gene expression related to intestinal barrier function in piglets with intrauterine growth retardation. Twenty-four litters of Landrace×Large White neonatal piglets were used in this study,and one IUGR piglet and one normal birth weight(NBW)piglet were selected from each litter. Eight IUGR piglets and eight NBW piglets were selected and slaughtered at 7,21 and 28 days of age,respectively,for small intestine sample collection and analysis.The results showed as follows:1)compared with the NBW piglets,the villus height in jejunum of IUGR piglets at 7 days of age and the villus height in jejunum and ileum at 21 days of age were significantly decreased(P<0.05),while the crypt depth in jejunum at 28 days of age was significantly increased(P < 0.05). 2)Compared with the NBW piglets,the mRNA relative expression levels of zonula occluden-1(ZO-1),claudin-1,occludin,interleukin-4(IL-4)and interleukin-8 in jejunal mucosa of IUGR piglets at 7 days of age and the mRNA relative expression levels of ZO-1 and occludin in jejunal mucosa and ZO-1 in ileal mucosa at21 days of age were significantly down-regulated(P<0.05);the mRNA relative expression levels of tumor necrosis factor-αin jejunal mucosa and interleukin-1βin ileal mucosa at 28 days of age were significantly upregulated(P<0.05),while the mRNA relative expression level of IL-4 in ileal mucosa was significantly downregulated(P<0.05). In summary,in IUGR piglets,jejunum and ileum morphology is impaired,tight junction protein gene expression is down-regulated,and immune cytokines expression is abnormal,which further affects intestinal barrier function.[Chinese Journal of Animal Nutrition,2022,34(2):1240-1246]
