摘要 研究飞燕草素(Delphinidin,Dp)抑制HER-2+乳腺癌细胞(MDA-MB-453)和HER-2-乳腺癌细胞(MCF-7)上皮细胞-间充质转化(EMT)效应。将不同浓度Dp分别处理MDA-MB-453和MCF-7细胞48 h,于倒置显微镜观察细胞形态及数量变化后,用蛋白质免疫印迹法和逆转录聚合酶链反应检测EMT标志性蛋白E-cadherin、Ncadherin、Vimentin、Fibronectin及关键调控因子锌指转录因子(Snail 1)表达。80μmol/L和100μmol/L的Dp能使MDA-MB-453和MCF-7乳腺癌细胞的E-cadherin标志蛋白表达上调,而降低与细胞迁移和侵袭能力有关的Ncadherin、Vimentin、Fibronectin及Snail因子表达水平。Dp体外具有抗MDA-MB-453和MCF-7乳腺癌EMT的效应,研究结果可为植物黄酮类化合物抗乳腺癌转移及辅助治疗提供新支撑。 In order to study the inhibitory effect on epithelial mesenchymal transition(EMT)of HER-2+breast cancer cells(MDA-MB-453)and HER-2-breast cancer cells(MCF-7)with delphinidin(Dp),MDA-MB-453 and MCF-7 cells were treated with different concentrations of Dp for 48 h,respectively.The morphological and quantitative changes of cells were observed by inverted microscope.The expression of EMT marker protein Ecadherin,N-cadherin,Vimentin,Fibronectin and the key regulatory factor zinc finger transcription factor(Snail 1)were detected by western blotting and reverse transcription polymerase chain reaction.The results showed that 80μmol/L and 100μmol/L Dp could increase the expression of E-cadherin marker protein in MDA-MB-453 and MCF-7 breast cancer cells,but reduce the expression level of N-cadherin,Vimentin,Fibronectin and Snail related to cell migration and invasion.Therefore,Dp had been proved to have the resistant effect on EMT of MDA-MB-453 and MCF-7 breast cancer cells in vitro.The research results could provide the new support for the anti metastasis and adjuvant therapy of plant flavonoids.
