摘要
目的探讨绿茶4种主要有效组分联合用于牙周炎大鼠治疗的效果。方法将288只牙周炎模型SD大鼠随机均分为空白组(生理盐水)、单用1组(茶多酚)、单用2组(咖啡碱)、单用3组(铁)、单用4组(锌)、联用1组(茶多酚联用铁)、联用2组(茶多酚联用锌)、联用3组(咖啡碱联用铁)、联用4组(咖啡碱联用锌),共9组,均干预5 d;比较9组大鼠干预前后龈沟液中脂联素(ADP)及内毒素(RSE)的含量,血清中肿瘤坏死因子-α(TNF-α)及白细胞介素-17(IL-17)含量,使用相加交互作用分析4个联用组对ADP的影响。结果干预后,ADP含量单用1组和单用2组高于空白组、联用1组高于单用1组、联用2组高于单用2组、联用4组高于单用2组,RSE含量正好相反(P<0.001);TNF-α含量,单用1组低于空白组、联用2组低于单用2组、联用4组低于单用2组(P<0.001);IL-17含量,4个单用组低于空白组,联用1组低于单用1组、联用2组低于单用2组、联用4组低于单用2组(P<0.001);4个联用组均存在相加交互作用,作用最强的是联用2组(AP=70.27)。结论绿茶中茶多酚和咖啡碱有抗菌作用,且茶多酚、咖啡碱、铁、锌兼有抗炎作用,其中茶多酚和锌搭配的使用效果最好。
Objective To explore the effect and difference of four main active components of green tea in the treatment of periodontitis in rats.Methods Two hundred and eithty-eight SD model rats with periodontitis were randomized into 9 groups:the Blank group(with saline),Single-use Group 1(with tea polyphenols),Single-use Group 2(with caffeine),Single-use Group 3(with iron),Single-use Group 4(with zinc),Combined-use Group 1(tea polyphenols combined with iron),Combined-use Group 2(tea polyphenols combined with zinc),Combined-use Group 3(caffeine combined with iron),Combined-use Group 4(caffeine combined with zinc),with 32 cases in each group.All the 9 groups of rats received intervention for 5 days.The contents of adipose tissue(ADP),reference standard endotoxin(RSE),tumor necrosis factor alpha(TNF-α),and interleukin-17(IL-17)before and after intervention in the 9 groups were detected and compared.The additive interactions were used to analyze the effects upon ADP in the four combined groups.Result After intervention,the contents of ADP in Single-use Group 1 and 2 were higher than in Blank group,they were higher in Combined-use Group 1 than in Single-use Group 1,higher in Combined-use Group 2 than in Single-use Group 2,and higher in Combined-use Group 4 than in Single-use Group 2(P<0.001),while the contents of RSE were just on the contrary(P<0.001).TNF contents of Single-use Group 1 was lower than in the Blank group,lower in Combined-use Group 2 than in Single-use Group 2;lower in Combined-use Group 4 than in Single-use Group 2(P<0.001).The contents of IL-17 in 4 groups of single use were lower than in the blank group,lower in Combined-use Group 1 than Single-use Group 1,lower in Combined-use Group 2 and Combined-use Group 4 than in Single-use Group 2,and the difference between the groups was statistically significant(P<0.001).There was additive interaction in all the 4 groups of combined use,and the strongest interaction was in Combined-use Group 2(AP=70.27).Conclusions The tea polyphenols and theine in green tea are anti-bacterial,while the tea polyphenols,theine,iron,and zinc also are also anti-inflammatory,and the combination of tea polyphenols and zinc possesses the best effect in the treatment of periodontitis in rats.
