阿斯青霉菌XK-12产铁载体特性及其抑菌活性 被引量:9

Characteristics of siderophores production by Penicilium asturianum XK-12 and its effect on antibacterial activity

摘要 对从健康草莓根际中分离出的一株阿斯青霉菌XK-12(Penicilium asturianum),进行了产铁载体的定性和定量分析,鉴定其所产铁载体为异肟羟酸型铁载体。并对XK-12产铁载体发酵条件进行了优化,利用其发酵液测定了铁载体对病原菌的抑制效果。结果表明,阿斯青霉菌XK-12产铁载体的最佳发酵条件为:3.0%NH_(4)Cl为氮源,2.0%葡萄糖为碳源,添加0.10%L-鸟氨酸盐酸盐,培养基中不含Fe^(3+),初始pH 6.0,温度28℃,250 ml培养瓶装液量为70 ml,发酵周期为168 h;在此条件下,XK-12产铁载体最高能达到0.173 mg/ml。在添加20%(体积分数)XK-12铁载体发酵液条件下,XK-12铁载体发酵液对草莓根腐病病原菌葡萄座腔菌(Neofusicoccum kwambonambiense XKD-1)的抑制率达到了100%。 The qualitative and quantitative analyses of siderophores production were carried out on Penicilium asturianum XK-12 isolated from healthy strawberry rhizosphere.The siderophores produced by XK-12 were identified as hydroxamic acid-type siderophores,and the conditions for producing siderophores were optimized.Finally,the fermentation broth of XK-12 was used to determine the inhibitory effect of siderophores on a pathogen.The results showed that the best fermentation conditions for siderophores production by XK-12 were as follows:3.0%NH_(4)Cl was used as nitrogen source,2.0%glucose was used as carbon source,0.10%of L-ornithine hydrochloride was added,there was no Fe^(3+)in the medium,the initial pH was 6.0,the temperature was 28℃,the liquid volume in 250 ml flask was 70 ml,and the fermentation period was 168 h.Under these conditions,0.173 mg/ml siderophores could be obtained.Under the condition of adding 20%(volume fraction)XK-12 fermentation broth,the inhibition rate of fermentation broth on strawberry root rot pathogen Neofusicoccum kwambonambiense was 100%.
