基于脂质组学的土鳖虫炮制品脂肪油含量 被引量:5

摘要 目的采用脂质组学方法研究干土鳖虫和甘草制土鳖虫两种饮片中的脂肪油含量。方法以石油醚为提取溶剂,采用连续回流方法提取土鳖虫焙干品和甘草制土鳖虫中的脂肪油,采用脂质组学技术,辅以SIMCA-P 14.0和SPSS 20.0软件,结合主成分分析(PCA)、正交偏最小二乘法分析(OPLS-DA)和Pearson相关性分析对土鳖虫两种不同炮制品中的脂质进行系统分析,并找出其差异成分。结果在土鳖虫干品中鉴定出85个脂质成分,其中甘油脂(GL)37个,脂肪酸(FA)37个,甘油磷脂(GP)10个,固醇脂(ST)1个。经PCA和OPLS-DA分析,结合VIP值和Pcorr的变量,以P<0.05为阈值,得出两种土鳖虫炮制品差异性脂质成分有68个,其中GL 32个,FA 26个,GP 9个,ST 1个。且多种脂质成分在两个炮制品内的相对含量不尽相同。结论不同炮制方法对土鳖虫中的脂质成分影响较大,这为不同土鳖虫炮制品的相关研究提供了基础。 Objective To systematically study the fatty oil content of dry Eupolyphaga sinensis products and licorice processing Eupolyphaga sinensis by the lipidomics research method.Methods Petroleum ether was used as the extraction solvent,and the continuous reflux method was used to extract the fatty oils of dry Eupolyphaga sinensis products and licorice processing Eupolyphaga sinensis products.Lipidomics technology was adopted,supplemented with SIMCA-P 14.0 and SPSS 20.0 related data software.Principal component analysis(PCA),orthogonal partial least squares analysis(OPLS-DA),and Pearson correlation analysis were used to systematically analyze the lipids in two different processed products of the soft-shelled beetle,and find out the different components.Results Eighty-five lipid components were identified in the dry products of the eupolyphaga sinensis,including 37 glycerolipids(GL),37 fatty acids(FA),10 glycerophospholipids(GP),and 1 sterol lipid(ST).After PCA and OPLS-DA analysis,combined with the VIP value and Pcorr variables,with P<0.05 as the threshold,it was concluded that there were 68 differential lipid components in the two kinds of eupolyphaga sinensis products,including 32 GL,26 FA,and 9 GP and 1 ST.And the relative content of various lipid components in the two processed products was not the same.Conclusion Different processing methods have a greater impact on the lipid composition of the eupolyphaga sinensis,which also provides a basis for related research on different processed products of the eupolyphaga sinensis.
