中晚期胃癌患者血清lncRNA-ANRIL和lncRNA H19表达及与预后相关性

Expression of serum lncRNA-ANRIL and lncRNA H19 in patients with advanced gastric cancer and their correlation with prognosis

目的探讨中晚期胃癌患者血清lncRNA-ANRIL和lncRNA H19表达及与预后相关性。方法选取浙江新安国际医院2017年1月至2019年1月收治的中晚期胃癌患者79例作为观察组;另选取同期本院健康体检者68例作为对照组。采用qRT-PCR法检测血清lncRNA-ANRIL和lncRNA H19相对表达量。所有患者通过门诊、电话方式随访,随访时间截止2021年1月,以患者死亡或至随访截止日为随访终点,统计平均总生存期(OS)。结果观察组血清lncRNA-ANRIL和lncRNA H19相对表达量均明显高于对照组(均P<0.01)。不同性别、年龄、BMI和肿瘤直径患者血清lncRNA-ANRIL相对表达量比较差异均无统计学(均P>0.05);而不同TNM分期及是否存在淋巴结转移患者血清lncRNA-ANRIL相对表达量比较差异均有统计学意义(均P<0.01)。不同性别、年龄、BMI和肿瘤直径患者血清lncRNA H19相对表达量比较差异均无统计学意义(均P>0.05);而不同TNM分期和是否存在淋巴结转移患者血清lncRNA H19相对表达量比较差异均有统计学意义(均P<0.01)。以中晚期胃癌患者血清lncRNA-ANRIL相对表达量中位数2.75为界,将患者分为lncRNA-ANRIL>2.75与≤2.75分别为43和36例,其OS分别为19和32个月,≤2.75患者总体生存情况明显优于>2.75患者(P<0.05)。以中晚期胃癌患者血清lncRNA H19相对表达量中位数1.83为界,将患者分为lncRNA H19>1.83与≤1.83分别为45和34例,其OS分别为21和31个月,≤1.83患者总体生存情况明显优于>1.83患者(P<0.05)。经多因素Cox回归模型分析,TNM分期、淋巴结转移、lncRNA-ANRIL和lncRNA H19相对表达量为影响预后独立危险因素(均P<0.05)。结论中晚期胃癌患者血清lncRNA-ANRIL和lncRNA H19呈高表达,且与预后密切相关。

Objective To investigate the expression of serum lncRNA-ANRIL and lncRNA H19 in patients with advanced gastric cancer and their correlation with prognosis.Methods Seventy nine patients with advanced gastric cancer admitted to Zhejiang Xin'an International Hospital from January 2017 to January 2019 and 68 healthy subjects(control group)were enrolled in the study.The expression of serum lncRNA-ANRIL and lncRNA H19 were detected with qRT-PCR.Patients were followed up the cases of death were documented and the overall survival(OS)was calculated.Results The relative expression of lncRNA-ANRIL and lncRNA H19 in gastric cancer patients were significantly higher than that in control group(P<0.05).There were no significant differences in relative expression of lncRNA-ANRIL and lncRNA H19 among patients with different gender,age,body mass index and tumor size(P>0.05),while there were significant differences among patients with different TNM staging and lymph node metastasis(P<0.05).The overall survival of patients with relative expression of lncRNA-ANRIL≤2.75 was better than relative expression of lncRNA-ANRIL>2.75(P<0.05),the overall survival of patients with relative expression of lncRNA H19≤1.83 was better than relative expression of lncRNA H19>1.83(P<0.05).Multivariate Cox regression model analysis showed that TNM staging,lymph node metastasis,lncRNA-ANRIL expression and lncRNA H19 expression were indepen-dent risk factors affecting prognosis of patients.Conclusion Serum lncRNA-ANRIL and lncRNA H19 are highly expressed in patients with advanced gastric cancer,and are closely related to the prognosis of patients.
