摘要
目的研究人参皂苷Rg5对宫颈癌HeLa细胞增殖、迁移、侵袭的影响以及作用机制。方法将HeLa细胞分为0μmol/L组、25μmol/L组、50μmol/L组、100μmol/L组;采用细胞计数(CCK-8)法检测不同浓度人参皂苷Rg5(0μmol/L、25μmol/L、50μmol/L、100μmol/L)对HeLa细胞增殖的影响;采用流式细胞术检测细胞周期和凋亡率;通过细胞划痕、Transwell实验检测各组细胞迁移、侵袭;采用蛋白质印迹法(Western blot)检测Notch神经同源蛋白1前体(Notch 1)信号通路蛋白水平的变化。结果与0μmol/L组相比,25μmol/L、50μmol/L、100μmol/L人参皂苷Rg5呈浓度依赖性抑制HeLa细胞增殖(P<0.05),促进凋亡(P<0.05),周期阻滞在G0/G1期(P<0.05),降低细胞迁移、侵袭能力(P<0.05),减少Notch 1、Jagged 1、兔抗发状分裂相关增强子1(Hes 1)蛋白水平(P<0.05)。结论人参皂苷Rg5可能通过调控Notch信号通路抑制HeLa细胞恶性生物学特性。
Objective To study the effects and mechanism of ginsenoside Rg5 on proliferation,migration and invasion of HeLa cells.Methods HeLa cells were divided into of 0μmol/L group,25μmol/L group,50μmol/L group,100μmol/L group and the proliferation were detected by cell counting kit-8(CCK-8).The cell cycle and apoptosis rate were observed by flow cytometry.The migration and invasion were detected by cell scratch test and Transwell.The protein levels of neurogenic locus notch homolog protein 1 precusor(Notch 1)signaling pathway were detected by Western blot assay.Results Compared with 0μmol/L group,ginsenoside Rg5 of 25μmol/L,50μmol/L,100μmol/L inhibited the proliferation of HeLa cells in a concentration-dependent manner(P<0.05),promoted apoptosis(P<0.05),inhibited cell cycle in G0/G1 phase(P<0.05),decreased cell migration and invasion(P<0.05),and reduced Notch 1,Jagged 1,hairy enhancer of split 1(Hes 1)protein levels(P<0.05).Conclusion Ginsenoside Rg5 may inhibit the biological characteristics of HeLa cells by regulating Notch signaling pathway.
