摘要
为探究萝芙木乙醇提取物(EERV)舒张小鼠气管平滑肌的作用机理,使用80 mmol·L^(-1) KCl或100μmol·L^(-1)乙酰胆碱(ACh)作为激动剂预刺激小鼠离体气管环,用生物机能实验系统检测了EERV对气管环张力的影响.用肺功能仪在活体水平上检测了EERV对小鼠呼吸系统阻力(Rrs)的影响.结果表明:EERV可通过影响VDCCs、NSCCs、NCX等离子通道来抑制外钙内流和内钙释放,降低胞内钙离子浓度,从而舒张由高钾或ACh诱导预收缩的气管平滑肌.EERV还可抑制ACh引起的Rrs升高.提示EERV有望成为用于治疗哮喘的新药.
To investigate the relaxation effect of ethanol extracts from Rauvofia verticillata(EERV)on mouse airway smooth muscle,the isolated tracheal rings was prestimulated with 80 mmol·L^(-1) KCl or 100μmol·L^(-1) ACh.Biological signal acquisition system was used to investigate the effect of EERV on tissue tension of isolated tracheal rings.At the living body level,pulmonary function meter was used to detect the effect of EERV on mouse respiratory system resistance(Rrs).The results showed that EERV could inhibit extracellular calcium influx and intracellular calcium release by inhibiting VDCCs,NSCCs,and NCX ion channels,reduce the concentration of intracellular calcium ion,and relax precontracted airway smooth muscle induced by high potassium or ACh.EERV could also inhibit the Rrs rise caused by ACh.It is suggested that EERV is a promising new drug for asthma.
