绵羊ZAP基因克隆与生物信息学分析

摘要 锌指抗病毒蛋白(ZAP)是一种重要的宿主限制性因子,能够抑制多种病毒的增殖。为研究绵羊ZAP(oZAP)的生物信息学特征和对该蛋白功能进行预测,本研究通过PCR扩增oZAP基因,其长度为2697 bp,编码898个氨基酸;生物信息学分析显示oZAP蛋白中丝氨酸(Ser)含量最高,半衰期为30 h,属于不稳定的亲水性蛋白;该蛋白不存在信号肽及跨膜结构,存在114个潜在的磷酸化位点和5个糖基化位点,二级结构预测中,参与形成无规则卷曲的氨基酸最多;功能结构域预测发现,该蛋白存在锌指域、WWE域和多聚核糖聚合酶功能域。本研究为进一步研究oZAP蛋白的抗病毒作用提供了有价值的信息。 Zinc fi nger antiviral protein is an important host restriction factor(ZAP)that inhibits the proliferation of a variety of viruses.In order to study the bioinformatic characteristics of sheep(Ovis aries)zinc fi nger antiviral protein(oZAP)and predict its function,oZAP gene was amplifi ed by PCR and sequenced.The oZAP gene was 2697 bp in length and encoded 898 amino acids.Bioinformatics analysis showed that oZAP protein was an unstable hydrophilic protein with the highest content of serine(Ser)with a half-life of 30 hours.The protein had no signal peptide and transmembrane structure but had 114 potential phosphorylation sites and 5 glycosylation sites.In the prediction of its secondary structure,most amino acids were involved in the formation of irregular curl.Functional domain prediction found that the protein contained zinc fi nger domain,WWE domain and polysulose polymerase domain.This study provided valuable information for further study on the antiviral effect of oZAP protein.
