Studies on Secondary Metabolites of the Deep-Sea Fungus Stachybotrys sp.3A00409 Under the Guidance of OSMAC Strategy

摘要 以OSMAC策略为指导,研究深海真菌Stachybotrys sp.3A00409的次级代谢产物多样性,对其燕麦大米培养基发酵产物中与原始培养基具有差异的部分进行靶向分离,共得到7个单体化合物,鉴定为丁内酯类化合物butyrolactone-Ⅰ(1)、butyrolactone-Ⅲ(2)、butyrolactone-Ⅱ(3)、2-O-methylbutyrolactone-Ⅱ(4)、萘类化合物methyl-6-acetyl-4-methoxy-5,7,8-trihydroxy-naphthalene-2-carboxylate(5)、吡喃酮衍生物pyrophen(6)和阿魏酸(7)。抗肿瘤实验结果显示化合物1,2,7对人口腔上皮癌KB细胞表现出了较强的增值抑制作用,IC_(50)在1.18~1.34μg∙mL^(-1)。 Under the guidance of OSMAC strategy,the diversity of secondary metabolites from deep-sea fungus Stachybotrys sp.3A00409 was explored,and the chemical composition and biological activity of fermentation products from rice and oat medium were studied.Seven compounds were isolated from the fermentation extract of rice and oat medium of Stachybotrys sp.3A00409,which were identified as butyrolactone-Ⅰ(1),butyrolactone-Ⅲ(2),butyrolactone-Ⅱ(3),2-O-methylbutyrolactone-Ⅱ(4),methyl-6-acetyl-4-methoxy-5,7,8-trihydroxy-naphthalene-2-carboxylate(5),pyrophen(6)and ferulic acid(7).Compound 1,2,7 showed proliferation inhibition against human oral epithelial carcinoma KB cells with IC_(50) values raging from 1.18 to 1.34μg∙mL^(-1).
