摘要 目的研究p21激活激酶4(PAK4)与RCC1和BTB结构域包含蛋白1(RCBTB1)的相互作用及其在胰腺癌细胞增殖过程中的作用。方法采用免疫共沉淀实验确定PAK4与RCBTB1在胰腺癌细胞内的相互作用;采用免疫荧光实验确定PAK4与RCBTB1在胰腺癌细胞内的定位和共定位位置;采用CCK-8实验探究过表达PAK4、RCBTB1对胰腺癌细胞增殖的影响;采用流式细胞术检测过表达PAK4、RCBTB1对胰腺癌细胞周期的影响;采用Western blotting检测过表达PAK4、RCBTB1对cyclin D1蛋白表达水平的影响。结果PAK4与RCBTB1在胰腺癌细胞内存在相互作用。PAK4与RCBTB1共定位于胰腺癌细胞核和核周细胞质内。过表达PAK4、RCBTB1促进胰腺癌细胞增殖,加速胰腺癌细胞的细胞周期进程,促进cyclin D1的表达。结论PAK4与RCBTB1相互作用促进胰腺癌细胞增殖。 Objective To explore the interaction between p21-activated kinase 4(PAK4)and RCC1 and BTB domain containing protein 1(RCBTB1),and its effects on the proliferation of pancreatic cancer cells.Methods An immunoprecipitation assay was used to determine the interaction between PAK4 and RCBTB1 in pancreatic cancer cells.An immunoconfocal assay was used to determine the localization and co-localization of PAK4 and RCBTB1 in pancreatic cancer cells.The CCK-8 assay was used to explore the effects of over-expressed PAK4 and RCBTB1 on the proliferation of pancreatic cancer cells.Flow cytometry was used to detect the effects of over-expressed PAK4 and RCBTB1 on cell cycle.Western blotting was used to detect the effect of over-expressed PAK4 and RCBTB1 on the expression of cyclin D1.Results Immunoprecipitation results indicated that PAK4 interacted with RCBTB1 in pancreatic cancer cells.Immunoconfocal assay results indicated the co-localization of PAK4 and RCBTB1 in the nucleus and perinuclear region of pancreatic cancer cells.The overexpression of PAK4 and RCBTB1 promoted cell proliferation,accelerated the cell cycle progression,and increased the expression of cyclin D1.Conclusion The interaction between PAK4 and RCBTB1 promotes the proliferation of pancreatic cancer cells.
