清血通脉饮对脂肪肝模型大鼠肝组织中CYP2E1、p38MAPK、CRP表达的影响 被引量:1

摘要 目的观察清血通脉饮对大鼠非酒精性脂肪肝模型的影响。方法将60只SD大鼠随机分为空白组、模型组、清血通脉饮(1.8 g/kg低剂量组,3.6 g/kg中剂量组,7.2 g/kg高剂量组),阿托伐他汀钙片对照组(10 mg/kg),除空白组外其余各组采用高脂饲料喂养大鼠8周,空白组、模型组灌胃给予蒸馏水,其余各组灌胃给与相应药物,给药容积(10 mL/kg),边造模边给药8周。实验结束后,检测各组大鼠血清谷丙氨酸转氨酶(GPT)、谷草转氨酶(GOT)的活性以及肝组织中的甘油三酯(TG)的含量,并采用苏木素-伊红(HE)染色观察肝组织脂肪变性损伤程度,采用免疫组化法检测肝组织中CYP2E1、p38 MAPK及CRP的表达。结果清血通脉饮可显著降低脂肪肝模型大鼠肝组织中TG的含量(P<0.0)。清血通脉饮可显著降低脂肪肝模型大鼠血清GPT、GOT的活性(P<0.05)。清血通脉饮可显著降低脂肪肝模型大鼠的肝细胞脂肪变性损伤程度。清血通脉饮可显著降低CYP2E1及CRP在脂肪肝模型大鼠肝组织中的表达(P<0.05)。清血通脉饮可显著增加p38 MAPK在脂肪肝模型大鼠肝组织中的表达(P<0.05)。结论研究结果表明清血通脉饮对高脂饮食诱发的脂肪肝及脂肪肝形成对肝组织的损害具有防治作用。 Objective To observe the effect of Qingxue Tongmai Yin(清血通脉饮)on non-alcoholic fatty liver model in rats.Method Sixty SD rats were randomly divided into a blank group,a model group,Qingxue Tongmai Yin groups(1.8 g/kg low-dose group,3.6 g/kg medium-dose group,7.2 g/kg high-dose group)and the atorvastatin calcium tablet control group(10 mg/kg).Except the blank group,rats in the other groups were fed with high-fat diet for 8 weeks.The blank group and the model group were orally administered with distilled water.The other groups were given the corresponding drugs by gavage with a volume of 10 mL/kg.The corresponding drugs were administered for 8 weeks while modeling.After the experiment,the activities of glutamic alanine aminotransferase(GPT),aspartate aminotransferase(GOT)in serum and the content of triglyceride(TG)in liver tissue of rats in each group were measured,and the fatty tissue damage of liver tissue was observed by HE staining.The expressions of CYP2 E1,p38 MAPK and CRP in liver tissue were detected by immunohistochemical method.Results Qingxue Tongmai Yin can significantly reduce the TG content in liver of fatty liver model rats(P<0.05).Qingxue Tongmai Yin can significantly reduce the activities of GPT and GOT in serum of fatty liver model rats(P<0.05).Qingxue Tongmai Yin can significantly reduce the degree of hepatic steatosis induced by high-fat diet.Qingxue Tongmai Yin can significantly reduce the expressions of CYP2 E1 and CRP in liver tissue of fatty liver model rats(P<0.05).Qingxue Tongmai Yin can significantly increase the expression of p38 MAPK in liver tissues of fatty liver model rats(P<0.05).Conclusion The results of this study indicate that Qingxue Tongmai Yin has a preventive and therapeutic effect on fatty liver and fatty liver damage induced by high-fat diet.
