核因子-κB p65和骨桥蛋白在子宫内膜癌组织与细胞中的表达及相关性研究 被引量:3

摘要 目的:探讨核因子-κB(NF-κB)p65和骨桥蛋白(OPN)在子宫内膜癌组织及细胞中的表达,并分析两者的相关性。方法:收集子宫内膜腺癌患者35例(子宫内膜腺癌组)、子宫内膜非典型增生患者35例(非典型增生内膜组)和正常子宫内膜患者35例(正常子宫内膜组),采用免疫组织化学法检测三组子宫内膜组织中NF-κB p65、OPN蛋白的表达。采用反转录PCR(RT-PCR)及Western blot检测正常内膜细胞及子宫内膜癌HEC-1-B细胞中两者蛋白及mRNA的相对表达水平。采用Pearson法分析NF-κB p65与OPN表达的相关性。结果:三组患者组NF-κB p65与OPN蛋白平均积分光密度(IOD)值比较差异有统计学意义(P<0.05)。子宫内膜腺癌组和非典型增生内膜组NF-κB p65与OPN蛋白IOD值高于正常子宫内膜组,且子宫内膜腺癌组高于非典型增生内膜组(均P<0.05)。Western blot和RT-PCR结果显示,NF-κB p65和OPN蛋白及mRNA在子宫内膜癌HEC-1-B细胞中的相对表达水平明显高于正常内膜细胞(均P<0.05)。Pearson相关性分析显示,子宫内膜腺癌组和非典型增生内膜组中,NF-κB p65和OPN蛋白表达呈正相关(r=0.78、0.88,均P<0.001),但在正常子宫内膜组中两者蛋白表达无相关性(P>0.05)。结论:NF-κB p65和OPN在子宫内膜癌组织及细胞中表达较高且呈正相关,有可能是形成该病的分子通路,为该病靶向治疗提供了理论依据及重要靶点。 Objective:To investigate the expression of nuclear factor kappa B(NF-κB)p65 and osteopontin(OPN)in endometrial carcinoma tissues and cells,and analyze the correlation between them.Methods:35 patients with endometrial adenocarcinoma(endometrial adenocarcinoma group),35 patients with endometrial atypical hyperplasia(atypical hyperplasia group)and 35 patients with normal endometrium(normal endometrium group)were collected.The expressions of NF-κB p65 and OPN in the endometrium of the three groups were detected by immunohistochemical method.RT-PCR and Western blot were used to detect the relative expression levels of their proteins and mRNAs in normal endometrial cells and endometrial cancer HEC-1-B cells.The correlation between NF-κB p65 and OPN expression was analyzed by Pearson method.Results:There was significant difference in average integrated optical density(IOD)of NF-κB p65 and OPN among three groups(P<0.05).The IOD values of NF-κB p65 and OPN in endometrial adenocarcinoma group and atypical hyperplasia group were higher than those in normal endometrium group,and endometrial adenocarcinoma group was higher than atypical hyperplasia group(all P<0.05).Western blot and RT-PCR results showed that the relative expression levels of NF-κB p65 and OPN proteins and mRNAs in endometrial carcinoma HEC-1-B cells were significantly higher than those in normal endometrial cells(all P<0.05).Pearson correlation analysis showed that there was a positive correlation between the expression of NF-κB p65 and OPN proteins in endometrial adenocarcinoma group and atypical hyperplasia group(r=0.78,0.88,all P<0.001),but there was no correlation in normal endometrium group(P>0.05).Conclusion:The expression of NF-κB p65 and OPN in endometrial cancer tissues and cells is high and positively correlated,which may be the molecular pathway of the disease,and provides a theoretical basis and important target for the targeted treatment.
