摘要
目的对清宣止咳颗粒进行止咳、祛痰与抗炎的药效学评价。方法建立小鼠氨水引咳模型、气管泌红模型与脂多糖气管滴注诱导的急性肺损伤模型对清宣止咳颗粒的止咳、祛痰、抗炎作用进行评价。观察各组小鼠的典型咳嗽动作,HE染色观察肺组织病理学改变,检测小鼠气管内酚红排泌量,急性肺损伤模型肺泡灌洗液中髓过氧化物酶(MPO)活性与白介素-1β(IL-1β)水平。结果在小鼠氨水引咳模型中,与空白组比较,模型组小鼠咳嗽次数增多,咳嗽潜伏期缩短;与模型组比较,清宣止咳颗粒1、2.5、5 g/kg组能有效抑制氨水引起的咳嗽(P<0.01),清宣止咳颗粒2.5、5 g/kg组对小鼠肺部病理改变均有显著改善。在气管泌红模型中,5 g/kg清宣止咳颗粒连续给药4 d能增加小鼠气管内酚红的排泌量(P<0.05)。在气管滴注脂多糖急性肺损伤模型中,清宣止咳颗粒能够抑制炎症引起的MPO、IL-1β水平升高(P<0.01)。结论清宣止咳颗粒可缓解刺激性气体所致的小鼠急性咳嗽并促进气管酚红分泌,减少急性肺损伤中炎症因子释放,具有显著的镇咳、祛痰、抗炎效果。
AIM To evaluate the pharmacodynamics of Qingxuan Zhike Granules in relieving cough,sputum and inflammation.METHODS The ammonia-induced mouse cough models,phenol red-induced tracheal excretion mouse models,and lipopolysaccharide(LPS) tracheal drip-induced acute lung injury mouse models were established for the assessment of antitussive,expectorant and anti-inflammatory effects of Qingxuan Zhike Granules.The mice were subjected to the observation of the typical cough action;the pathological changes of lung tissue by HE staining;and the determinations of the amount of tracheal phenol red excretion,and MPO activity,and IL-1β level in alveolar lavage fluid of the acute lung injury models respectively.RESULTS The ammonia-induced cough models displayed,increased frequency of coughs and shortened incubation period of cough in contrast to the mice of the blank group.Compared with the model group,Qingxuan Zhike Granules groups(dosed with 1,2.5,5 g/kg each time) shared effectively inhibited ammonia-induced cough(P<0.01);Qingxuan Zhike Granules groups given dosages of 2.5,5 g/kg were observed with improvement in the pathological changes of mouse lungs.To the tracheal models of phenol red excretion,the administration of 5 g/kg Qingxuan Zhike Granules for four consecutive days increased the excretion of phenol red(P<0.05).Upon the acute lung injury models induced by tracheal LPS drip,Qingxuan Zhike Granules inhibited the increase of MPO and IL-1β due to inflammation(P<0.01).CONCLUSION Qingxuan Zhike Granules exhibit significant antitussive,expectorant and anti-inflammatory effects because of their efficacy in relieving acute cough caused by irritating gas,promoting tracheal phenol red secretion,and reducing the release of inflammatory factors in acute lung injury in mice.
