miR-103b在妊娠期糖尿病孕妇外周血、脐血中的表达及对糖代谢指标的影响 被引量:1

Effect of miR-103b expression in peripheral blood and umbilical cord blood on glucose metabolism in pregnant women with gestational diabetes

摘要 目的本研究旨在探讨miR-103b在妊娠期糖尿病(gestational diabetes mellitus,GDM)孕妇外周血、脐血中的表达水平及对糖代谢指标的影响。方法选取2018年1月至2021年1月于沧州市人民医院住院分娩的175例GDM孕妇作为研究对象(GDM孕妇组),并纳入同期在本院分娩的180例健康孕妇作为健康对照组。收集孕妇分娩前外周血和分娩后脐带血,记录一般资料和糖代谢指标。采用实时定量聚合酶链反应检测外周血和脐血中miR-103b和分泌型卷曲相关蛋白4(secreted frizzled-related protein 4,SFRP4)mRNA表达量。结果GDM孕妇组的新生儿体重、空腹血糖(fasting plasma glucose,FPG)、空腹胰岛素(fasting insulin,FINS)、胰岛素抵抗指数(homeostasis model assessment-insulin resistance,HOMA-IR)和糖化血红蛋白(hemoglobin A1c,HbA1c)水平均显著高于健康对照组(P均<0.05)。GDM孕妇组外周血中miR-103b相对表达量显著低于健康对照组的表达量(t=5.324,P<0.001);GDM孕妇组外周血中SFRP4 mRNA相对表达量显著高于健康对照组的表达量(t=4.836,P<0.001)。GDM孕妇组新生儿脐血中miR-103b相对表达量显著低于健康对照组的表达量(t=4.724,P<0.001);新生儿脐血中SFRP4 mRNA相对表达量显著高于健康对照组的表达量(t=4.862,P<0.001)。GDM孕妇组外周血中miR-103b表达量与新生儿体重、FPG、FINS、HOMA-IR和HbA1c均呈显著负相关关系(P均<0.05);新生儿脐血中miR-103b表达量与新生儿体重、FINS、HOMA-IR和HbA1c均呈显著负相关关系(P均<0.05)。GDM孕妇组外周血和新生儿脐血中miR-103b表达水平的Pearson相关性分析结果显示两者呈显著正相关关系(r=0.716,P<0.001)。结论miR-103b在GDM孕妇外周血、脐血中的表达显著降低,且具有一致性,可能参与到GDM的糖代谢调控机制中。 Objective To investigate the effect of miR-103b expression in peripheral blood and umbilical cord blood on glucose metabolism in pregnant women with gestational diabetes mellitus(GDM).Method A total of 175 GDM women who delivered in Cangzhou People􀆳s Hospital from January 2018 to January 2021 were enrolled in GDM group,and 180 healthy pregnant women who delivered in our hospital during the same period were included as the control group.The peripheral blood sample before delivery and umbilical cord blood sample after delivery were collected,and the general data and glucose metabolism parameters were recorded.The mRNA expression levels of miR-103b and secreted frizzled-related protein 4(SFRP4)in peripheral blood and umbilical cord blood were detected by quantitative real-time polymerase chain reaction.Result The neonatal body weight(BW),fasting plasma glucose(FPG),FINS,HOMA-IR and hemoglobin A1c(HbA1c)in GDM group were significantly higher than those in control group(all P<0.05).The expression level of miR-103b in peripheral blood(t=5.324,P<0.001)and umbilical cord blood(t=4.724,P<0.001)in GDM group was significantly lower than that of control group.The expression level of SFRP4 mRNA in peripheral blood(t=4.836,P<0.001)and umbilical cord blood(t=4.862,P<0.001)in GDM group was significantly higher than that of control group.In GDM women,the expression level of miR-103b in peripheral blood of was negatively correlated with neonatal BW,FPG,FINS,HOMA-IR and HbA1c(all P<0.05).The expression level of miR-103b in umbilical cord blood was also negatively correlated with neonatal BW,FINS,HOMA-IR and HbA1c(all P<0.05).Pearson correlation analysis showed that the miR-103b expression levels in peripheral blood was significant positively correlated with the miR-103b expression levels in umbilical cord blood(r=0.716,P<0.001).Conclusion The expression of miR-103b in peripheral blood and umbilical cord blood in GDM women may decrease and be consistent,which may be involved in the regulation of glucose metabolism in GDM.
