摘要
目的:观察强心汤对心梗后心衰大鼠CaN-NFAT3信号通路的影响,探讨其治疗心梗后心衰大鼠的作用机制。方法:将64只SD雄性大鼠随机分为对照组10只,模型组、缬沙坦组、强心汤组各18只,除对照组大鼠行假手术外,其余各组制备慢性心衰大鼠模型。8周后,给予对应药物灌胃治疗4周。采用PowerLab/4SP生物信号处理分析系统测定大鼠血流动力学[左心室收缩压(left ventricular systolic pressure,LVSP)、左室舒张末压(left ventricular end-diastolic pressure,LVEDP)、左室压力上升和下降变化最大速度的参数值(±LVdp/dtmax)];采用酶联免疫吸附法分别测定大鼠血浆血管紧张素Ⅱ(plasma angiotensinⅡ,AngⅡ)、血清钙调磷酸酶(calcineurin,CaN)水平;采用Western blot法检测大鼠心肌活化T细胞核因子(nuclear factor activated T3,NFAT3)蛋白表达量。结果:与模型组比较,缬沙坦组、强心汤组LVEDP水平降低(P<0.05),缬沙坦组+LVdp/dtmax、-LVdp/dtmax水平降低(P<0.01),强心汤组LVSP、+LVdp/dtmax及-LVdp/dtmax水平均升高(P<0.05)。与模型组比较,强心汤组、缬沙坦组大鼠血浆AngⅡ、血清CaN、心肌NFAT3蛋白表达水平均明显下降(P<0.01),且强心汤组上述指标下调程度均大于缬沙坦组(P<0.01)。结论:强心汤能够有效地改善心衰大鼠的心功能,其机制可能与影响CaN-NFAT3信号通路相关。
Objective:To observe the effects of Qiangxin decoction on CaN-NFAT3 signaling pathway in heart failure rats after myocardial infarction,and explore therapeutic mechanism of treating heart failure rats after myocardial infarction.Methods:All 64 SD male rats were randomized into ten rats in the control group,18 rats in the model group,valsartan group and Qiangxin decoction group,except the rats in the control group performed sham operation,the other groups were prepared into rat models with chronic heart failure.After eight weeks,the rats were drenched with the corresponding medicines for four consecutive weeks.Hemodynamic indexes[LVSP,LVEDP and(±LVdp/dtmax)]of the rats were detected by using PowerLab/4SP biological signaling processing and analysis system;the contents of AngII and CaN of the rats were measured by using ELISA method;the expressions of NFAT3 protein in cardiac muscle of the rats were detected by usign Western blot method.Results:Compared with the model group,the levels of LVEDP in valsartan group and selfmade Qiangxin decoction group lowered(P<0.05),the levels of+LVdp/dtmax and-LVdp/dtmax lowered in valsartan group(P<0.01),while the levels of LVSP,+LVdp/dtmax and-LVdp/dtmax increased in selfmade Qiangxin decoction group(P<0.05).Compared with the model group,the expressions of plasma AngII,serum CaN and myocardial NFAT3 protein in self-made Qiangxin decoction group and valsartan group reduced notably(P<0.01),and the decrease of these indexes in self-made Qiangxin decoction group were better than these of valsartan group(P<0.01).Conclusion:Self-made Qiangxin decoction could effectively improve cardiac function in heart failure rats,and its mechanism might be associated with improving CaN-NFAT3 signaling pathway.
