摘要
目的探究长链非编码RNA(lncRNA)P21、lncRNA H19在妊娠期糖尿病(GDM)患者血清和胎盘组织中表达及临床意义。方法回顾性选取2019年1月到2021年1月在大连市妇女儿童医疗中心(集团)定期产检且分娩的73例GDM孕妇作为GDM组,另选41名同期健康孕妇作为对照组。将GDM组患者按照病情严重程度分为轻度组(n=42)、中重度组(n=31)。收集受试者临床资料,比较各组孕妇血清及胎盘组织中lncRNA P21、lncRNA H19表达水平,分析其与胰岛素抵抗指数(HOMA-IR)及新生儿体重关系。结果GDM组血清和胎盘组织中lncRNA P21、lncRNA H19表达水平均高于对照组,差异均有统计学意义(P<0.05)。中重度组GDM患者血清和胎盘组织中lncRNA P21、lncRNA H19表达水平、HOMA-IR、新生儿体重高于轻度组GDM患者,差异均有统计学意义(P<0.05)。Pearson相关性分析显示,血清和胎盘组织中lncRNA P21、lncRNA H19表达与HOMA-IR、新生儿体重呈正相关(P<0.05),而血清、胎盘组织中lncRNA P21与lncRNA H19表达均呈正相关(P<0.05)。结论lncRNA P21、lncRNA H19在GDM患者血清和胎盘组织中表达均上调,两者可能共同参与了GDM进展,且与新生儿出生体重升高有关。
Objective To explore the expression and clinical significance of long non-coding RNA(lncRNA)P21 and lncRNA H19 in serum and placenta tissue of patients with gestational diabetes(GDM).Methods A total of 73 pregnant women with GDM who had undergone regular check-ups in Dalian Municipal Women and Children's Medical Center from January 2019 to January 2021 were selected as the GDM group,and 41 healthy pregnant women during the same period were selected as the control group.Patients in GDM group were divided into mild group(n=42)and moderate-severe group(n=31)according to the severity of their illness.The clinical data of the subjects were collected,and the expression levels of lncRNA P21 and lncRNA H19 in the serum and placental tissues of the two groups of pregnant women were compared.The relation with the insulin resistance index(HOMA-IR)and newborn weight was analyzed.Results The expression levels of lncRNA P21 and lncRNA H19 in serum and placental tissues of the GDM group were higher than those in the control group,the differences were statistically significant(P<0.05).The expression levels of lncRNA P21,lncRNA H19,HOMA-IR,and neonatal weight in serum and placental tissues of patients with moderate to severe GDM were higher than those of patients with mild GDM,and the differences were statistically significant(P<0.05).Pearson correlation analysis showed that the expression of lncRNA P21 and lncRNA H19 in serum and placenta tissue was positively correlated with HOMA-IR and newborn weight,the differences were statistically significant(P<0.05),while the expression of lncRNA P21 and lncRNA H19 in serum and placenta tissue were positively correlated(P<0.05).Conclusion The expressions of lncRNA P21 and lncRNA H19 are up-regulated in the serum and placenta tissues of GDM patients.Both may involve in the progression of GDM and cause the weight increase of newborns by increasing insulin resistance.
