摘要
非酒精性脂肪肝病(non-alcoholic fatty liver disease, NAFLD)作为高发病率的慢性肝病之一,其主要病理表现为除酒精等其他明确因素所致的肝脂质蓄积,大量研究表明某些编码脂类代谢相关蛋白质的基因的多态性与肝内脂肪蓄积、变性等NAFLD的病理生理学特征相关,本研究就游离脂肪酸摄取及转运、肝脏脂肪合成、脂肪酸β-氧化及极低密度脂蛋白(very low density lipo protein, VLDL)合成及分泌等脂类代谢不同环节综述了相关基因多态性在NAFLD中的作用,以期从基因组水平为发现预防、诊断和治疗NAFLD的新思路提供参考。
Non-alcoholic fatty liver disease(NAFLD), one of the chronic liver diseases with high morbidity, is characterized by pathological hepatic lipid accumulation not attributable to alcohol or other definite causes. Numerous studies have shown that polymorphisms in certain genes which encode proteins involved in lipid metabolism are associated with fat accumulation and degeneration in the liver. In order to provide references for the new ideas of the prevention, diagnosis and therapy of NAFLD from the genomic level, this review discusses the role of gene polymorphisms in lipid metabolism that regulate different aspects of lipid metabolism, including free fatty acid uptake and transport, hepatic lipid synthesis, fatty acid β-oxidation and very low density lipoprotein synthesis and secretion.
