山西不同产地酸枣遗传多样性与化学成分关联性分析 被引量:3

Correlation Analysis Between Chemical Components and Genetic Diversity of Ziziphus jujuba var. spinosa from Different Areas in Shanxi Province

摘要 目的探究山西不同产地酸枣简单重复序列区间(ISSR)分子标记遗传多样性与5种主要化学成分之间的关系。方法利用ISSR分子标记技术对山西省26个不同产地酸枣进行遗传多样性分析,并对药材酸枣仁中5种化学成分进行含量测定,在此基础上使用TASSEL软件对ISSR分子标记实验结果和化学成分含量进行关联性分析。结果山西不同产地酸枣具有丰富的遗传多样性,遗传一致度在0.727 2~0.949 8之间,聚类分析表明地理位置并非影响酸枣居群间基因交流的主要因素。不同产地酸枣5种化学成分含量范围:木兰花碱含量0.745~1.537 mg·g^(-1)、斯皮诺素含量0.358~0.815 mg·g^(-1)、6^(''')-阿魏酰斯皮诺素含量0.122~0.571 mg·g^(-1)、酸枣仁皂苷A含量0.265~0.424 mg·g^(-1)、酸枣仁皂苷B含量0.100~0.226 mg·g^(-1)。群体遗传结构分析将26个产地酸枣分为3个类群,不同地理来源的群体间存在基因交流,与聚类分析结果基本一致,基于GLM模型分析发现,在P<0.05水平上,12条引物总共有19个位点与5种化学成分含量发生关联,各位点对表型变异的解释率在15.35%~33.87%。结论酸枣仁中5种化学成分及关联位点、基因调控的研究,可为山西道地药材酸枣品种选育、物种保护、资源综合利用提供有利参考。 OBJECTIVE To reveal the relationship between inter-simple sequence repeat(ISSR) genetic diversity of Ziziphus jujuba var. spinosa from different areas of Shanxi and five main chemical components. METHODS The genetic diversity of Ziziphus jujuba var. spinosa from 26 different areas in Shanxi province was analyzed by ISSR molecular markers, and the contents of five chemical components in Ziziphi Spinosae Semen(ZSS) were determined. On this basis, Tassel software was used to analyze the correlation between the results of ISSR molecular marker experiment and the content of chemical components. RESULTS The genetic diversity of Ziziphus jujuba var. spinosa from different areas in Shanxi was rich, and the genetic agreement is 0.727 2-0.949 8. Cluster analysis showed that geographic location was not the main factor affecting gene exchange among populations. The contents of five chemical components in ZSS from different areas were as follows: magnoflorine 0.745-1.537 mg·g^(-1), spinosin 0.358-0.815 mg·g^(-1), 6^(''')-feruloyl spinosin 0.122-0.571 mg·g^(-1), jujuboside A 0.265-0.424 mg·g^(-1), jujuboside B 0.100-0.226 mg·g^(-1). The genetic structure analysis of 26 different areas of Ziziphus jujuba var. spinosa was divided into three groups. There was gene exchange among different geographical sources, which was consistent with the cluster analysis results. Based on GLM model analysis, 19 loci of 12 primers were associated with 5 chemical components content in total at P<0.05 level. The explanation rate of phenotypic variation was 15.35%-33.87% among all the points. CONCLUSION The results provide a basis for the study of the five chemical components, associated sites and gene regulation of ZSS, and provide a favorable reference for the breeding, species protection and comprehensive utilization of Ziziphus jujuba var. spinosa in Shanxi province.
