摘要
目的基于网络药理学分析麻黄-苦杏仁、辛夷-苍耳子药对治疗儿童过敏性鼻炎哮喘综合征(CARAS)的多成分、多靶点、多途径作用机制。方法结合文献报道,利用TCMSP数据库筛选出麻黄、苦杏仁、辛夷、苍耳子的化学成分和潜在靶标;利用GeneCards、OMIM数据库获取CARAS的疾病靶点;通过R软件获取药物-疾病交集靶基因;利用String数据库构建蛋白互作(PPI)网络图并筛选出核心靶点,利用R语言对核心靶基因进行GO富集分析和KEGG通路分析;利用Sytoscape 3.7.2软件可视化化合物-交集靶点网络、关键通路-关键靶点网络。结果从麻黄-苦杏仁、辛夷-苍耳子药对中筛选得到槲皮素、山萘酚、木犀草素等52个活性成分,作用于MAPK1、MAPK3、AKT1、JUN等24个儿童过敏性鼻炎哮喘综合征核心靶点,参与调节DNA结合转录因子活性、活性氧、脂多糖等生物过程,调控AGE-RAGE信号通路、TNF信号通路、IL-17等信号通路。结论揭示了麻黄-苦杏仁、辛夷-苍耳子药对多成分、多靶点、多通路治疗CARAS的作用机制。
Objective Based on the network pharmacology,to analyze the multi-component,multi-target and multi-channel mechanism of action of Ephedra-Armeniacae semen amarum and Magnoliae flos-Xanthii fructus herb pairs in the treatment of combined allergic rhinitis and asthma syndrome in children.Methods Combined with literature reports,the chemical components and potential targets of Ephedra,Armeniacae semen amarum,Magnoliae flos and and Xanthii fructus were screened out by TCMSP database;the disease targets of Caras were obtained by GeneCards and OMIM database;the drug disease intersection target genes were obtained by R software;the protein interaction(PPI)network map was constructed by using string database and the core targets were screened out by R language to get Go enrichment analysis and KEGG pathway analysis;compound intersection target network and key pathway key target network were visualized by sytoscape 3.7.2 software.Results 52 active components,such as quercetin,kaempferol and luteolin were screened from Ephedra-Armeniacae semen amarum and Magnoliae flos-Xanthii fructus.They act on 24 core targets of combined allergic rhinitis and asthma syndrome,such as MAPK1,MAPK3,AKT1 and JUN,participate in the biological process of regulating DNA binding transcription factor activity,active oxygen and lipopolysaccharide,and regulate AGE-RAGE signal pathway,TNF signal pathway and IL-17 and other signal pathways.Conclusion The mechanism of action of Ephedra-Armeniacae semen amarum and Magnoliae flos-Xanthii fructus herb pairs on multi-component,multi-target and multi-channel treatment of Caras was revealed.
