摘要
目的研究lncRNA-AC005062.1对结直肠癌细胞侵袭及迁移的影响并研究其机制。方法实时荧光定量PCR(qRT-PCR)检测新鲜结直肠癌及癌旁正常组织中lncRNA-AC005062.1表达差异,通过siRNA转染结直肠癌细胞HT29,敲低lncRNA-AC005062.1表达量,qRT-PCR验证转染效率。其次,通过划痕实验检测HT29细胞迁移变化;Transwell实验检测HT29细胞侵袭变化;Western blot检测上皮间质转化(EMT)相关分子(E-Cadherin、N-Cadherin、Vimentin)的表达量改变。结果与癌旁正常组织相比,结直肠癌组织中lncRNA-AC005062.1表达量明显增高(P<0.05),敲低HT29细胞lncRNA-AC005062.1表达量后,细胞侵袭及迁移能力明显降低(P<0.05),细胞中的E-Cadherin蛋白表达量明显增高(P<0.05),而N-Cadherin及Vimentin表达量明显降低(P<0.05)。结论lncRNA-AC005062.1在结直肠癌中表达明显升高,并且影响HT29细胞侵袭及迁移能力,这种改变可能与E-Cadherin、N-Cadherin、Vimentin的表达及逆转EMT进程有一定相关性。
Objective To investigate the effect of lncRNA-AC005062.1 on the invasion and migration of colorectal cancer cells and its mechanism.Methods The difference of lncRNA-AC005062.1 expression in fresh colorectal cancer and adjacent normal tissues was detected by real-time quantitative fluorescence PCR(qRT-PCR).The expression of lncRNA-AC005062.1 was knocked down by siRNA transfection in colorectal cancer cells HT29,and the transfection efficiency was verified by qRT-PCR.Secondly,the migration of HT29 cells was detected by scratch test.HT29 cell invasion was detected by Transwell assay.The expression of EMT-related molecules(E-cadherin,N-cadherin,Vimentin)was detected by Western blot.Results Compared with the adjacent normal tissues,the expression of lncRNA-AC005062.1 in colorectal cancer tissues was significantly increased(P<0.05).The invasion and migration ability of HT29 cells were significantly decreased after the expression of lncRNA-AC005062.1 was knocked down(P<0.05).The expression of E-cadherin protein was significantly increased(P<0.05),while the expression of N-cadherin and Vimentin were significantly decreased(P<0.05).Conclusion The expression of lncRNA-AC005062.1 is significantly increased in colorectal cancer and affects the invasion and migration ability of HT29 cells,which may be related to the expression of E-cadherin,N-cadherin and Vimentin and the reversal of EMT process.
