摘要
为了对采集自3只德国牧羊犬小肠内的复孔绦虫进行种类鉴定和遗传进化分析,通过形态学方法对分离到的虫体进行孔雀绿染色后显微镜观察,并进一步基于线粒体部分基因(p cox 1和p nad 1)进行PCR扩增和序列分析。显微镜观察结果表明,分离到的虫体形态和内部结构符合犬复孔绦虫的特征。通过对扩增的靶基因序列及其遗传进化分析发现,3个虫体样本的p cox 1和p nad 1序列完全一致,构建系统发生树显示这些虫体均与Dipylidium caninum(AB732959)位于同一进化支。以上结果提示,分离到的绦虫种为犬复孔绦虫(D.caninum)。研究结果为宠物复孔绦虫病的诊断和防控奠定了基础。
The aims of the present study were to identify and genetically analyze Dipylidium caninum collected from the small intestine of German shepherd dog.Adult cestodes were firstly identified based on the morphological observation under microscopy after staining method,and then the part genes of cox 1 and nad 1 were analyzed by PCR and sequencing.The results showed that the external and internal morphological features of parasites are consistent with that of D.caninum.Sequencing and genetic analysis showed that the sequences of p cox1 and p nad1 from three samples are the same,and they are located in the same clade with D.caninum in phylogenetic tree based on sequences of p cox1 and p nad1.These results indicated that the cestode in the present study has high identity to D.caninum,suggesting that the cestode collected from German shepherd dog is D.caninum.The findings in this study will provide theoretical basis for diagnosis,control and prevention of Dipylidiasis in pets.
