茯苓多糖对磷酸铵镁致羊肾小管上皮细胞损伤的保护作用 被引量:4

Protective effects of Poria cocos polysaccharide extract on injury of sheep renal tubular epithelial cells induced by magnesium ammonium phosphate

摘要 氧化损伤在结石的形成过程中具有重要作用。茯苓多糖(PCP)具有抗炎、抗氧化、免疫调节等多种作用。本研究探讨PCP对磷酸铵镁(MAP)诱导的羊肾小管上皮细胞(RTECs)炎症和氧化应激的影响。结果显示,PCP显著减轻MAP诱导的氧化应激,表现为乳酸脱氢酶(LDH)和丙二醛(MDA)水平降低,超氧化物歧化酶(SOD)活性升高。PCP抑制MAP对炎症细胞因子IL-6和TNF-α的诱导作用。Western blot试验显示,PCP能增加Nrf2、 HO-1和NQO1的表达。综上所述,PCP通过提高细胞Nrf2、NQO1、HO-1的表达水平,从而对MAP诱导的肾小管上皮细胞氧化应激和炎症起到保护作用,这为羊尿结石的中药防控提供了依据。 Oxidative damage plays an important role in the formation of calculus.Poria cocos polysaccharide(PCP) has the effects of anti-inflammatory, anti-oxidation, immune regulation etc.In this study, the effects of PCP on inflammation and oxidative stress in sheep renal tubular epithelial cells(RTECs) induced by magnesium ammonium phosphate(MAP) was investigated.The results showed that PCP significantly alleviated MAP induced oxidative stress by decreasing the levels of lactate dehydrogenase(LDH) and malondialdehyde(MDA) and increasing the activity of superoxide dismutase(SOD).PCP inhibited the induction of IL-6 and TNF-α by MAP.By Western blot, it showed that PCP could increase the expressions of Nrf2,HO-1 and NQO1.In conclusion, by increasing the expression levels of Nrf2,NQO1 and HO-1,PCP can protect RTECs from oxidative stress and inflammation induced by MAP,which provides the basis for the prevention and control of sheep urinary calculi with traditional Chinese medicine.
