猪睾丸组织的形态学观察、细胞凋亡和巴豆酰化修饰

Morphological observation,apoptosis and crotonylation modification of porcine testis tissue

摘要 制作睾丸组织切片,运用HE染色和细胞TUNEL染色分别观察不同发育阶段猪睾丸的形态学和细胞凋亡;提取睾丸组织蛋白,利用免疫印迹检测不同发育阶段猪睾丸蛋白的Kcr修饰水平;最后利用免疫荧光技术检测Kcr蛋白在猪睾丸组织中的定位。结果显示,随着月龄的增长,猪睾丸精细管管腔直径增加,睾丸组织在4月龄时精细胞开始分裂,凋亡细胞开始显著增加;睾丸细胞中有大量蛋白发生Kcr修饰,大部分蛋白的Kcr修饰水平会随着月龄的增加而减少;免疫荧光结果显示在猪睾丸间质细胞、足细胞和生精细胞中都能检测到Kcr蛋白定位。试验表明,在猪睾丸发育过程中,4月龄时精细胞开始分裂,凋亡细胞显著增加,大部分蛋白Kcr修饰水平有下降趋势且主要定位在睾丸体细胞和生精细胞中。 Boar testis at different developmental stages were obtained,fixed,dehydrated,embedded and cut.The morphology and apoptosis of testicular tissues were observed by HE staining and TUNEL staining,respectively.Western blot was used to assess the levels of lysine crotonylation modification in boar testis.Immunofluorescence was used to determine the location of the crotonylated protein in boar testis.The results showed that the lumen diameter of seminiferous tubule enlarged with increasing age.Spermatogonium in testicular tissues began to divide and apoptosis cells began to increase significantly at four months old.A large number of proteins in testicular tissues undergone crotonylation modification and the level of crotonylation modification of most proteins decreases with increasing age.Immunofluorescence results demonstrated that crotonylated proteins mainly localized in leydig cells,sertoli cells and spermatogenic cells.It was shown that during testicular development,spermatogonium began to divide,apoptosis cells increased significantly at four months old and most of proteins crotonylation modification show a downward trend and crotonylated proteins were mainly localized in somatic cell and spermatogenic cell in boar testis.
