摘要
目的:观察电针长强穴对缺血缺氧脑损伤(HIBD)大鼠学习记忆能力及海马PI3K、Akt蛋白表达的影响。方法:制备HIBD大鼠,随机分为电针长强组(长强组)、电针非穴组(非穴组)、模型组,各8只,另随机选取并制备假手术组大鼠8只。长强组取长强穴,电针刺激(连续波,频率为2 Hz,强度为2 mA)20 min,连续治疗10 d为1个疗程,共1个疗程;非穴组取大鼠右胁肋下缘一横指处,余操作同长强组;假手术组和模型组在相同环境下饲养,不予干预。治疗结束第2天予Morris水迷宫试验,取脑组织,应用免疫印迹技术观察大鼠海马PI3K、Akt蛋白的表达。结果:与假手术组比较,模型组逃避潜伏期显著延长(P<0.01),目标象限游泳时间百分比显著下降(P<0.01);与模型组比较,长强组逃避潜伏期显著缩短(P<0.05),目标象限游泳时间百分比显著上升(P<0.05)。与假手术组比较,模型组PI3K、Akt蛋白表达显著降低(P<0.01);与模型组比较,长强组PI3K、Akt蛋白表达显著升高(P<0.01,P<0.05)。结论:电针长强穴可能通过激活海马PI3K及Akt蛋白的表达,对神经元异常发育进行调节,进而改善其学习记忆能力。
Objective:To observe the effects of electroacupuncture at Changqiang(GV 1)point on learning-memory ability and the expression of PI3 K and Akt protein in hippocampus of neonatal rats with hypoxic-ischemic brain damage(HIBD).Methods:Eight rats were randomly divided into the electroacupuncture long-strength group(chang-strength group),the electroacupuncture non-acupoint group(non-acupoint group)and model group,and eight rats were randomly selected and prepared from the sham operation group.In the Changqiang group,changqiang acupoint was selected,electroacupuncture stimulation(continuous wave,frequency:2 Hz,intensity:2 mA)was performed for 20 min,and continuous treatment for 10 days was one course of treatment,a total of one course of treatment.The lower margin of right rib was taken from the non-acupoint group,and the remaining operation was the same as that of the changqiang group.The sham operation group and the model group were raised in the same environment without intervention.On the second day after the treatment,Morris water maze test was performed.Brain tissue was taken and immunoblotting technique was used to observe the expressions of PI3 K and Akt proteins in the hippocampus of rats.Results:Compared with control group,model group escape latency significantly prolonged(P<0.01),target quadrant swimming percentage significantly decreased(P<0.01);Compared with model group,strong escape latency was significantly shortened(P<0.05),target quadrant swimming time percentage significantly increased(P<0.05).Compared with control group,model group of PI3 K,and Akt protein expression significantly reduced(P<0.01);Compared with model group,long strong group of PI3 K,Akt protein expression increased(P<0.01,P<0.05).Conclusion:Electroacupuncture of Changqiang may by activating PI3 K and Akt protein expression of the hippocampus,adjusted to the abnormal development of neurons,and improve the learning-memory ability.
