摘要
目的:以氧嗪酸钾(OA)建立高尿酸血症大鼠模型,研究矢志方对不同病程高尿酸血症大鼠氧化应激损伤的改善作用。方法:160只SD大鼠分为4组:正常组、模型组、矢志方组、别嘌醇组,每组40只,以750 mg·kg^(-1)·d^(-1)OA氧嗪酸钾灌胃建立高尿酸血症大鼠模型,以矢志方或者别嘌醇作为干预药物,实验第3、5、7、11周末各组取10只大鼠检测ROS、线粒体ROS、MDA、SOD、NO、GSH-Px、CAT水平,PCR、Western Blot及免疫组化法检测过氧化产物3-NT、4-HNE mRNA和蛋白的表达。结果:与模型组比较,矢志方组可降低不同病程高尿酸血症大鼠ROS及线粒体ROS水平,增加抗氧化系统SOD、NO、GSH-Px、CAT的浓度,降低过氧化系统MDA浓度,抑制3-NT、4-HNE mRNA和蛋白的表达。结论:矢志方可改善不同病程阶段的高尿酸血症大鼠肾脏氧化应激损伤。
Objective:Hyperuricemic rat model was established by using potassium oxonate(OA)to study the ameliorative effect of Shizhifang on oxidative stress injury in rats with different course of hyperuricemia.Methods:A total of 160 SD rats were divided into 4 groups:normol group,model group,Shizhifang group,allopurinol group,40 rats in each group,750 mg·kg^(-1)·d^(-1)OA was administered to establish a hyperuricemia rat model,and Shizhifang or allopurinol was used as an intervention drug in the experiment.At the end of the 3 rd,5 th,7 th,and 11 th week,10 rats from each group were executed to detect the levels of ROS,mitochondrial ROS,MDA,SOD,NO,GSH-Px,and CAT.PCR,Western Blot and immunohistochemical methods were used to detect the mRNA and protein expression of peroxide product 3-NT and 4-HNE.Results:Compared with the model group,Shizhifang group could reduce the levels of ROS and mitochondrial ROS in hyperuricemic rats with different courses,increase the concentration of antioxidants of SOD,NO,GSH-Px,CAT,reduce the concentration of peroxidation system MDA,and inhibit the mRNA and protein expression of 3-NT and 4-HNE.Conclusion:Shizhifang can improve kidney oxidative stress injury in rats with different stages of hyperuricemia.
