摘要
目的:建立基于标准汤剂的槟榔、焦槟榔配方颗粒HPLC特征图谱并进行鉴别。方法:采用高效液相色谱法,色谱柱为Agilent ZORBAX Eclipse XDB-C_(18)(4.6mm×250mm,5μm),流动相为乙腈-0.1%磷酸溶液梯度洗脱,流速为1.0mL·min^(-1),柱温为30℃,检测波长为215nm;以相对保留时间评价特征图谱,并根据特征峰峰面积比进行鉴别。结果:HPLC特征图谱均确定了4个共有峰,并指认了两个成分(槟榔碱和儿茶素);根据其中共有峰的峰面积比进行二者的鉴别。结论:首次建立了槟榔、焦槟榔标准汤剂及配方颗粒的HPLC特征图谱。该方法的精密度、重复性、稳定性、耐用性良好,可鉴别槟榔、焦槟榔标准汤剂及配方颗粒,对槟榔、焦槟榔配方颗粒的鉴别及质量控制具有指导意义和参考价值。
Objective:To establish the quality evaluation system of Areca Catechuand Charred A.Catechuformula granules based on the quality study of standard decoction.Methods:The HPLC characteristic chromatogram was obtained using a Agilent ZORBAX Eclipse XDB-C_(18)(4.6 mm×250 mm,5μm)column.The mobile phase consisted of acetonitrile and 0.1% phosphoric acid solution and eluted in a gradient mode at the flow rate of 1.0 mL·min^(-1).The column temperatureand the detection wavelength was set at 30℃ and 215 nm,respectively.Relative retention time were used to evaluate the characteristic atlas,and the characteristic atlas peak-to-peak area ratio was used for identification.Results:There were 4 common peaks in the HPLC characteristic chromatogram of 18 batches of Areca Catechuand Charred A.Catechuformula granules,and 2 common component(arecoline and catechin)were identified.According to the peak area ratio of the common peaks,the two are identified.Conclusion:The characteristic chromatogram is precision,stable and reliable.The HPLC characteristic chromatogram of standard decoction and formula granules of Areca Catechu and Charred A.Catechu was established for the first time.It can also be used to identify the standard decoction and formula particles of Areca Catechu and Charred A.Catechu,which has guiding significance and reference value for the identification and quality control of the formula particles of Areca Catechu and Charred A.Catechu.
