摘要
建立了检测苹果幼果中根皮苷含量的二极管阵列-高效液相色谱法。色谱条件为C_(18) (250 mm×4.6 mm,5μm),柱温30℃,流动相为水+乙睛=70+30,检测波长为275 nm,标准曲线的线性范围为14.36~287.2μg/mL,相关系数R^(2)=0.999 9,检出限为1.16 mg/kg,定量限为3.83 mg/kg,回收率范围为97.84%~99.30%,平均回收率为98.48%。精密度S_(RSD)为0.87%,精密度良好。利用该方法分析了富士、秦阳、秦冠、红星、红盖露、中秋王、金世纪、黄元帅、洼里红、短枝10种苹果幼果中根皮苷的含量。结果表明不同品种根皮苷含量有很大区别。叶片的含量比果肉和果皮都高。洼里红的叶片中含量最高,它的果肉和果皮中也有较高的含量。红星果肉和果皮中含量最高,金世纪叶片、果皮和果肉含量最低。
A method to detect phloridzin in young apple fruit was established using high performance liquid chromatography(HPLC) with a diode array detector (DAD).Chromatographic separation was conducted on a C_(18) (250 mm×4.6 mm,particle size 5μm) with an elution using a mobile phase made up of water and acetonitrile as at 275 nm.The method had a linear range from 14.36-287.2μg/mL with correlation coefficient of 0.999 9,the limit of detection was 1.16 mg/kg and the limit of quantification was 3.83 mg/kg.The average recovery was 98.48%ranging from 97.84%to 99.30%.The relative retention time (S_(RSD)) of precision was 1.67%.Phloridzin in young apple fruits from 10 different varieties was determined by the HPLC method.The results showed that the contents of phloridzin in were quite different.Phloridzin in leaves was the highest.The variety of Walihong was the highest content of phloridzin in leaves and higher in pulp and peel.With the highest contents in pulp and peel,Starkrim zon performed well.The contents of leaf,peel and pulp of Jinshiji were the lowest.
