转录因子WRKY71对拟南芥根系发育的影响 被引量:1

Effects of transcription factor WRKY71 on root development of Arabidopsis thaliana

摘要 WRKY家族是拟南芥中最大的转录因子家族之一,为探究其中WRKY71在拟南芥根系生长发育过程中的调控作用,本研究利用WRKY71突变体exb1、过表达株系exb1-D及WRKY71-EAR融合子株系EXB1-SRDX,通过对其幼苗表型分析发现,突变体与野生型无明显差异,WRKY71过表达株系萌发率显著降低了50.6%,WRKY71-EAR融合子株系根毛平均数量、根毛平均长度、主根平均直径及平均鲜重比野生型分别增加了4.6%、118.4%、1.8%、16%,而过表达株系则呈相反趋势,分别显著减少了50.2%、48.5%、17.8%、17.6%。以上结果表明突变体中仍然存在与WRKY71功能冗余的同源基因,WRKY71过表达抑制幼苗早期发育,进而阻碍植株生物量累积,而WRKY71-EAR融合子则能够显著增加拟南芥幼苗早期的生物量。本文为进一步研究WRKY71及同源基因的生物学功能奠定了重要基础。 The WRKY family is one of the largest transcription factor families in Arabidopsis.In order to explore the regulatory role of WRKY71 in the growth and development of Arabidopsis root,the plants of WRKY71 mutant exb1,over-expression and WRKY71-EAR fusion lines were subjected to phenotype analysis.It was found that there was no significant difference between the mutant and the wild type,while the germination rate of the WRKY71 overexpression line was significantly reduced by 50.6%.Compared to wild type,the average number of root hair,average length of root hair,average diameter of primary root and average fresh weight of seedling from WRKY71-EAR plants were increased by 4.6%,118.4%,1.8%,and 16%plant respectively,while the over-expression line showed the opposite results,which was significantly reduced by 50.2%,48.5%,17.8%,and 17.6%respectively.The above results indicated that there were still function-redundant homologous genes with WRKY71 in the mutant.Over-expression of WRKY71 inhibited the early development of seedlings,thereby reduced the accumulation of plant biomass,while the WRKY71-EAR fusion could significantly increase the early biomass of Arabidopsis seedlings.This article provided an important foundation for further research on the biological functions of WRKY71 and homologous genes.
