摘要
本研究以‘章姬’草莓为试验材料,通过外植体消毒、高温处理剥离茎尖、愈伤组织诱导、不定芽诱导、生根培养建立‘章姬’草莓茎尖脱毒及快繁体系。‘章姬’草莓茎尖脱毒以38℃高温连续处理38 d为宜;最佳的灭菌处理为75%酒精60s,2%次氯酸钠8 min;茎尖愈伤组织诱导培养基为MS+6-BA 1.5 mg·L^(-1)+IBA 0.75 mg·L^(-1);不定芽诱导培养基为MS+6-BA1.5 mg·L^(-1)+NAA 0.5 mg·L^(-1),最佳生根培养基为1/2 MS+NAA 0.75 mg·L^(-1)。建立的茎尖脱毒及快繁体系为‘章姬’草莓的开发利用以及工厂化育苗提供参考。
In this study,the stem tip virus-free and rapid propagation system of‘Zhangji’strawberry was established by explants disinfection,high temperature treatment for exfoliation of stem tip,callus induction,adventitious bud induction and rooting culture.The optimal treatment for detoxification of stem tip of‘Zhangji’strawberry was 38℃for 38 days.The best combination of sterilization treatment was 75%alcohol 60 s,2%sodium hypochlorite 8 min.Shoot tip callus induction medium was MS+6-BA1.5 mg·L^(-1)+IBA 0.75 mg·L^(-1),the medium for adventitious bud induction was MS+6-BA 1.5 mg·L^(-1)+NAA 0.5 mg·L^(-1),and the best rooting medium was 1/2 MS+NAA 0.75 mg·L^(-1);The establishment of shoot tip virus-free and rapid propagation system can provide reference for the development and utilization of‘Zhangji’strawberry and industrial vegetative reproduction.
