高压氧对去卵巢大鼠骨微结构改善及骨代谢的影响机制探究

摘要 目的研究高压氧对去卵巢大鼠骨微结构改善及骨代谢的影响机制。方法SPF级雌性Wistar大鼠共30只,按照随机数字法分为空白组、模型组、高压氧组共三组,每组各10只。空白组不做任何处理,模型组、高压氧组大鼠均去卵巢法建立骨质疏松模型大鼠。三组均正常饲养,其中高压氧组每天高压氧干预30 min。4周后酶联免疫法分析各组大鼠血清骨代谢标志物及雌二醇表达的变化;CT影像分析各组大鼠股骨远端骨密度水平及骨小梁微结构,并进行计量学分析。结果与空白组相比,模型组大鼠骨密度明显降低,大鼠骨代谢标志物骨特异性碱性磷酸酶(BALP)、核结合因子-α1(CBF-α1)、Ⅰ型胶原羧基末端肽(CTX-Ⅰ)、Ⅰ型前胶原氨基端前肽(PⅠNP)、骨钙素(OC)及雌二醇含量减少,差异有统计学意义(P<0.05);股骨远端相对骨体积分数(BV/TV)、骨小梁数量(Tb.N)、骨小梁厚度(Tb.Th)、连接密度(Conn.D)、骨小梁分离度(Tb.Sp)、结构模型指数(SMI)改变,差异有统计学意义(P<0.05)。高压氧干预4周后,与模型组对比,高压氧组大鼠骨密度、骨小梁微结构以及骨代谢标志物明显增加,骨强度及弹性显著增加,差异有统计学意义(P<0.05)。结论高压氧可以在一定程度上提高骨质疏松模型大鼠骨代谢水平,增加骨含量,加固骨小梁微结构,可作为绝经后骨质疏松患者的治疗手段之一。 Objective To investigate the mechanism of the effect of hyperbaric oxygen on the improvement of bone microstructure and bone metabolism in de-ovalized rats.Methods A total of 30 female Wistar rats of SPF grade were included in this study and divided into 3 groups according to the random number method,namely,the blank group,the model group and the hyperbaric oxygen group,with 10 rats in each group.The rats in the blank group were not treated,and the rats in the model group and the hyperbaric oxygen group were de-ovalized to establish osteoporosis model rats.Three groups of rats were raised normally,and the rats in the hyperbaric oxygen group were intervened with hyperbaric oxygen for 30 min per day.Four weeks later,the changes of serum bone metabolism markers and estradiol expression in each group were analyzed by enzyme-linked immunosorbent assay.The bone density level of the distal femur and bone trabeculae microstructure in each group of rats were analyzed by CT imaging,and metrological analysis was performed.Results Compared with the blank group,the bone density of rats in the model group was significantly lower,and the levels of bone metabolic markers,such as bone-specific alkaline phosphatase(BALP),core-binding factor-α1(CBF-α1),cross-linked C-telopeptide of type I collagen(CTX-I),N-terminal propeptide of typeⅠprocollagen(PⅠNP),osteocalcin(OC)and estradiol,were changed,with statistically significant differences(P<0.05).Compared with the blank group,the relative bone volume fraction(BV/TV),trabecular number(Tb.N),trabecular thickness(Tb.Th),joint density(Conn.D),trabecular separation(Tb.Sp),and structural model index(SMI)of the distal femur were reduced in the model group,with statistically significant differences(P<0.05).After 4-week intervention with hyperbaric oxygen,compared with the model group,the rats in the hyperbaric oxygen group showed a significant increase in bone density,trabecular microstructure and bone metabolism markers,and a significant improvement in bone strength and elasticity,with statistically significant differences(P<0.05).Conclusion Hyperbaric oxygen can improve bone metabolism level,increase bone content and reinforce trabecular microstructure in osteoporosis model rats to a certain extent,and can be used as one of the treatments for postmenopausal pat ients with osteoporosis.
