基于网络药理学探讨茵陈蒿汤治疗溶血性黄疸的作用机制 被引量:1

摘要 目的:探讨茵陈蒿汤治疗溶血性黄疸的作用机制。方法:在TCMSP数据库筛选茵陈蒿汤活性成分,利用Pub Chem数据库收集活性成分2D结构图并在Swiss Target Prediction数据库预测相关靶点,在Gene Cards数据库预测溶血性黄疸靶点,二者靶点构建韦恩图得到共同作用靶点。利用STRING数据库、Cytoscape软件构建共同靶点蛋白质-蛋白质相互作用网络图并得到Degree值。在DAVID数据库进行基因本体论(GO)、京都基因与基因组百科全书(KEGG)分析,并利用Cytoscape软件构建靶点-通路网络图。结果:本研究收集到茵陈蒿汤活性成分41个,活性成分相关靶点1511个,溶血性黄疸靶点312个,共同靶点37个,以肿瘤坏死因子(Tumor Necrosis Factor,TNF)、白细胞介素-6(Interleukin 6,IL-6)、白细胞介素-2(Interleukin 2,IL-2)、信号转导因子和转录激活因子3(Signal Transducer and Activator of Transcription 3,STAT3)等相互作用最明显,共同靶点以多种方式参与调控炎症反应、细胞因子介导的信号通路、STAT3蛋白酪氨酸磷酸化的正调控等120个生物过程,并参与调节Toll样受体、TNF、细胞凋亡等53个信号通路。结论:茵陈蒿汤可能通过调控炎症反应、细胞凋亡等机制发挥治疗溶血性黄疸的作用。 Objective:To explore the mechanism of the Yinchenhao decoction(茵陈蒿汤)in the treatment of hemolytic jaundice.Methods:Screen the active ingredients of the Yinchenhao decoction in the TCMSP database,collect the 2D structure diagram of the active ingredients in the PubChem database and predict the relevant targets in the Swiss Target Prediction database,predict the hemolytic jaundice target in the GeneCards database,and the common targets were obtained because of the Venn diagram constructed by the two targets.Use STRING database and Cytoscape software to construct a common target PPI network diagram and get the Degree value.The GO and KEGG analysis were performed on the DAVID database,and the target-pathway network diagram was constructed using Cytoscape software.Results:This study collected 41 active ingredients of the Yinchenhao decoction,1511 targets related to active ingredients,312 targets for hemolytic jaundice,and 37 common targets.Among them,TNF,IL-6,IL-2,STAT3,etc.have the most obvious interaction.The common target was involved in 120 biological processes such as the regulation of inflammation,cytokine-mediated signaling pathways,and positive regulation of STAT3 protein tyrosine phosphorylation in a variety of ways,as well as the regulation of Toll-like receptors,TNF,apoptosis,etc.53 signal pathways.Conclusion:The Yinchenhao decoction may play a role in the treatment of hemolytic jaundice by regulating inflammation,cell apoptosis and other mechanisms.
